【摘要】：目的利用光纖倏逝波生物傳感器適合現(xiàn)場快速檢測的優(yōu)勢,初步建立1型嗜肺軍團菌的快速檢測平臺。方法優(yōu)化最佳檢測條件,并在此條件下確定檢測Lp1的靈敏度和特異性,同時通過檢測人工模擬樣品確認該方法檢測實際樣本的靈敏度和特異性。結果該方法 40 min之內(nèi)自動完成檢測。捕獲抗體與目標物最佳反應時間、反應體系與檢測抗體的最佳反應時間均為10 min,捕獲抗體最佳使用濃度是100μg/ml,檢測抗體最佳使用濃度為20μg/ml。在此條件下得到該方法檢測Lp1的靈敏度為1.8×103cfu/ml,檢測的特異性強。結論光纖倏逝波生物傳感器快速檢測方法為大樣本量Lp1污染嚴重水樣的現(xiàn)場快速篩查提供一種新途徑。
[Abstract]:Objective to establish a platform for rapid detection of Legionella pneumophila type 1 by using optical fiber evanescent wave biosensor. Methods the optimal detection conditions were optimized, and the sensitivity and specificity of Lp1 detection were determined under these conditions, and the sensitivity and specificity of the method were confirmed by detecting artificial simulated samples. Results the detection was completed automatically within 40 min of the method. The optimal reaction time for the capture antibody and the target, the optimal reaction time for the reaction system and the detection of the antibody were both 10 min,. The optimal concentration of the capture antibody was 100 渭 g / ml, and the best concentration for detection of the antibody was 20 渭 g / ml. Under these conditions, the sensitivity of this method for detecting Lp1 is 1.8 脳 10 ~ 3 cfur / ml, and the detection specificity is strong. Conclusion the fast detection method of fiber optic evanescent wave biosensor provides a new way for rapid screening of large sample size Lp1 polluted water samples.
