模擬失重條件下骨形態(tài)發(fā)生蛋白-2對(duì)大鼠骨肉瘤成骨樣細(xì)胞基因表達(dá)的影響

【摘要】:目的觀察回轉(zhuǎn)器模擬失重條件下骨形態(tài)發(fā)生蛋白 2 (BMP 2 )對(duì)大鼠骨肉瘤成骨樣細(xì)胞 (ROS1 7/ 2 .8)基因表達(dá)的影響。方法在 1G和回轉(zhuǎn)器模擬失重條件下對(duì)ROS1 7/ 2 .8細(xì)胞進(jìn)行培養(yǎng) ,培養(yǎng)液中含5 0 0ng/ml的BMP 2。在實(shí)驗(yàn)的 2 4、48和 72h提取細(xì)胞總RNA ,進(jìn)行反轉(zhuǎn)錄PCR ,檢測(cè)Ⅰ型膠原α1鏈(COL Ⅰα1 )及堿性磷酸酶 (ALP)mRNA的表達(dá) ,并計(jì)算與內(nèi)參照 β -肌動(dòng)蛋白 ( β actin)mRNA水平的比值。結(jié)果BMP 2誘導(dǎo)組的COL Ⅰα1mRNA表達(dá)在 2 4h和 48h顯著高于無(wú)BMP 2組 (P 0 .0 1 ) ,ALPmRNA水平在 48h和 72h顯著高于無(wú)BMP 2組 (分別為P 0 .0 1 ,P 0 .0 5 ) ;BMP 2誘導(dǎo) 48h后模擬失重組的COL Ⅰα1mRNA水平顯著低于 1G對(duì)照組 (P 0 .0 1 ) ,模擬失重 48h和 72h后ALPmRNA表達(dá)顯著低于 1G對(duì)照組 (P 0 .0 1 )。結(jié)論BMP 2可促進(jìn)大鼠骨肉瘤成骨樣細(xì)胞的分化相關(guān)基因的表達(dá) ,在模擬失重條件下細(xì)胞對(duì)BMP 2的促分化作用的反應(yīng)性下降。
[Abstract]:Objective to observe the effect of bone morphogenetic protein-2 (BMP2) on the expression of ROS17 / 2 路8) in osteosarcoma like cells of rats under simulated weightlessness.
