局部海水浸泡對SMIR模型大鼠疼痛行為學和繼發(fā)神經病理改變的影響

【摘要】:目的評價局部海水浸泡對皮膚/肌肉切口牽拉痛(SMIR)模型大鼠疼痛行為學及繼發(fā)神經病理改變的影響。方法選擇健康成年雄性SD大鼠48只,隨機分為模型對照組(MC組,只建立SMIR模型,不予浸泡處理)、生理鹽水浸泡組(NS組,建立模型后局部傷口給予生理鹽水浸泡1h)、海水浸泡組(SW組,建立模型后局部傷口給予海水浸泡1h),于術前1d及術后1、3、5、7、14、21、28d測定機械縮足反射閾值(MWT),于術后3、28d各組分別隨機取8只大鼠,取術側隱神經行HE染色、取術側L3及L4背根神經節(jié)應用電鏡技術觀察其超微結構變化。結果 SW組術后各時點MWT值均明顯低于NS組和MC組,且至術后28d仍明顯低于術前基礎水平;NT組和NS組術后各時點MWT值無明顯差異,兩組術后28d的MWT值與術前基礎水平相當。SW組術后3、28d隱神經HE染色顯示均有不同程度的神經損傷表現(xiàn),而MC組和NS組未見異常;術后3、28d電鏡觀察見SW組背根神經節(jié)神經元內有大量空泡樣線粒體和自噬小體,MC組和NS組未見明顯病理改變。結論海水浸泡局部傷口可加重SMIR模型大鼠的急性疼痛,并可能導致其疼痛向慢性化轉變,海水浸泡導致的外周神經損傷、背根神經節(jié)神經元超微結構改變可能參與了其疼痛行為學改變的機制。
[Abstract]:Objective to evaluate the effects of local seawater immersion on pain behavior and secondary neuropathological changes in skin / muscle incision stretch pain rats. Methods 48 healthy adult male SD rats were randomly divided into model control group and control group. The SMIR model was established only. The saline immersion group was treated with NS group. After the model was established, the local wound was given saline immersion for 1 hour, and the seawater immersion group was given the SW group. After the model was established, the local wound was treated with seawater immersion for 1 hour. The mechanical foot reflex threshold (MWTT) was measured at 1 day before operation and 1 day after operation. Eight rats were randomly selected from each group on the 3rd and 28th days after operation. The saphenous nerve of the operative side was stained with HE. The ultrastructural changes of L3 and L4 dorsal root ganglion were observed by electron microscopy. Results the MWT values in SW group were significantly lower than those in NS group and MC group at all time points after operation. At 28 days after operation, the MWT values of NT group and NS group were still significantly lower than those of preoperative basic level NT group and NS group, and there was no significant difference at each time point between NT group and NS group. The MWT values of the two groups on the 28th day after operation were similar to the basic level before operation. The HE staining of the saphenous nerve in the SW group showed different degrees of nerve injury on the 38th day after operation, but there was no abnormality in the MC group and NS group. A large number of vacuolated mitochondria and autophagy bodies in the neurons of dorsal root ganglion neurons in group SW were observed by electron microscope on the 328th day after operation. There were no obvious pathological changes in MC group and NS group. Conclusion the acute pain of SMIR model rats can be aggravated by seawater immersion in local wound. It may lead to the transformation of pain to chronic, the peripheral nerve injury caused by seawater immersion, and the ultrastructural changes of dorsal root ganglion neurons may be involved in the mechanism of pain behavior changes.
