運(yùn)動(dòng)對(duì)不同濃度2,3,7,8-TCDD染毒大鼠附睪功能的影響

【摘要】:目的:選SD大鼠為實(shí)驗(yàn)對(duì)象,研究運(yùn)動(dòng)對(duì)不同劑量2,3,7,8-TCDD染毒大鼠生殖系統(tǒng)的影響,探討運(yùn)動(dòng)干預(yù)對(duì)2,3,7,8-TCDD的生殖毒性是否有削減作用。方法:將100只8周齡雄性SD大鼠隨機(jī)分為10組,分別為EC/ET1/ET2/ET3/ET4,NC/NT1/NT2/NT3/NT4。T1(ET1,NT1),T2(ET2,NT2),T3(ET3,NT3),T4(ET4,NT4)組大鼠的首次注射劑量分別為0.4 μ g/kg ·BW,1.6μ g/kg·BW,6.4μg/kg·BW和25.6μ g/kg·BW,之后每周均給予上述劑量的21%作為染毒持續(xù)劑量。C(EC,NC)組大鼠注射染毒劑量為0μ g/kg·BW,根據(jù)體重比例注射等量玉米油。E組大鼠運(yùn)動(dòng)方案為大鼠尾部負(fù)重5%自身體重,每次游泳運(yùn)動(dòng)30min,每周5天,共持續(xù)8周。N組大鼠靜養(yǎng)8周。8周后,將所有大鼠處死,測(cè)定大鼠體重,附睪重量,附睪臟器質(zhì)量指數(shù),大鼠血清睪酮(T)、黃體生成素(LH)、卵泡刺激素(FSH)。結(jié)果:1.E組大鼠體重非常顯著性小于N組(P0.01)。C組顯著大于T2、T3、T4組,T1組顯著大于T3、T4組,T2、T3組顯著大于T4組(P0.01)。2.T4組附睪臟器指數(shù)顯著大于其余4組,其余四組見間差異不顯著。3.N組FSH顯著小于E組(P0.01),劑量主效應(yīng)不顯著,運(yùn)動(dòng)與否和劑量的交互效應(yīng)不顯著。4.運(yùn)動(dòng)主效應(yīng)顯著:N組(安靜組)的LH顯著小于E組(運(yùn)動(dòng)組);劑量主效應(yīng)顯著:T1組顯著大于其他各組,T2組顯著大于T4組,其余各組間差異不顯著。運(yùn)動(dòng)與否和劑量的交互作用顯著。N組各劑量水平間LH無顯著性差異,但E組上T1組顯著大于C組和T4組,其余各組間差異不顯著。5.運(yùn)動(dòng)與否和劑量的交互作用對(duì)T的影響效應(yīng)顯著,T2水平上N組(安靜組)顯著大于E組(運(yùn)動(dòng)組)。結(jié)論:1.隨著TCDD染毒劑量增加,大鼠正常生長(zhǎng)發(fā)育受抑制的程度更加明顯,劑量效應(yīng)關(guān)系明顯。2.TCDD染毒擾亂了下丘腦-垂體-性腺軸的正常功能,使此軸上的激素水平異常改變。3.TCDD與游泳訓(xùn)練具有拮抗性,游泳可適度減輕TCDD中毒癥狀,但只在染毒濃度不超過25.6μg/kg·BW時(shí)有效。4.TCDD開始明顯在機(jī)體中發(fā)揮生殖毒性的"閾值"濃度大概在0.4~1.6μg/kg·BW之間。
[Abstract]:Objective: to study the effects of exercise on the reproductive system of rats exposed to different doses of TCDD and to explore whether exercise intervention can reduce the reproductive toxicity of TCDD. Methods: 100 8-week-old male SD rats were randomly divided into 10 groups: EC/ET1/ET2/ET3/ET4,NC/NT1/NT2/NT3/NT4.T1 (ET1,NT1), T2 (ET2,NT2), T3 (ET3,NT3) and T4 (ET4,). The initial injection doses of NT4 group were 0.4 渭 g/kg BW,1.6 渭 g/kg BW,6.4 渭 g/kg BW and 25.6 渭 g/kg BW, respectively. After that, 21% of the above doses were given weekly as the continuous dose of. C (EC,NC). The rats in the group were injected with 0 渭 g/kg BW,. According to the proportion of body weight, the rats in group E were injected with the same amount of corn oil. The rats in group E carried 5% body weight on the tail of the rats, swimming for 30 minutes, 5 days a week for a total of 8 weeks. The rats in group N were treated with rest for 8 weeks. After 8 weeks, all the rats were killed. Body weight, epididymal weight, organ mass index of epididymis and serum testosterone (T), luteinizing hormone (LH),) follicle stimulating hormone (FSH).) were measured in rats. Results: 1. The body weight of group E was significantly lower than that of group N (P0.01). C group was significantly larger than that of T2T3 T4 group, T1 group was significantly larger than T3 T4 group, T2 group. Group T3 was significantly larger than group T4 (P0.01). The organ index of epididymis in 2.T4 group was significantly higher than that in other four groups, but there was no significant difference among the other four groups. 3.The FSH in group N was significantly lower than that in group E (P0.01), and the main dose effect was not significant. The interaction between exercise and dose was not significant. 4. The LH of group N (quiet group) was significantly lower than that of group E (exercise group), and the dose effect was significant: group T1 was significantly larger than other groups, group T2 was significantly larger than group T4, and there was no significant difference among the other groups. The interaction between exercise and dose was significant. There was no significant difference in LH between different dose levels in group N. however, the LH in group E was significantly higher than that in group C and group T4, but there was no significant difference among the other groups. The interaction between exercise and dose had a significant effect on T. Group N (quiet group) was significantly greater than group E (exercise group) at T2 level. Conclusion: 1. With the increase of the dose of TCDD, the inhibition of normal growth and development of rats was more obvious, and the dose-effect relationship was obvious. 2.TCDD treatment disturbed the normal function of hypothalamus-pituitary-gonadal axis. 3.TCDD has antagonistic effect with swimming training. Swimming can alleviate the symptoms of TCDD poisoning moderately. But only when the concentration of 4.TCDD was not more than 25.6 渭 g/kg BW, the threshold concentration of 4.TCDD to produce reproductive toxicity was about 0.4 渭 g/kg BW or 1.6 渭 g/kg BW.
