MST過表達及亞砷酸鈉對SH-SY5Y細胞的影響

目的:探討亞砷酸鈉(Na As O2)、巰基丙酮酸硫轉(zhuǎn)移酶(MST)基因過表達對SH-SY5Y細胞的作用及機制。方法:將空載體轉(zhuǎn)染細胞、MST過表達細胞各分為對照組、Na As O2組、TUDCA(內(nèi)質(zhì)網(wǎng)應(yīng)激阻斷劑)組、Na As O2聯(lián)合TUDCA組,采用MTT、Western blot和酶活性檢測等方法觀察細胞活力、Bax、Bcl-2凋亡相關(guān)蛋白表達及Caspase-3活性。結(jié)果:50μmo L/L Na As O2暴露24 h顯著降低空轉(zhuǎn)組細胞活力和Bcl-2蛋白水平(P <0.01),增加Bax蛋白表達及Caspase-3活性(P <0.01); TUDCA預處理可以逆轉(zhuǎn)以上變化,Bcl-2的蛋白表達則在MST基因過表達細胞砷暴露后發(fā)生顯著增加,但可被TUDCA所拮抗(P <0.01)。結(jié)論:內(nèi)質(zhì)網(wǎng)應(yīng)激可能參與砷、內(nèi)源性H2S對SH-SY5Y細胞損傷的影響。
