【摘要】：背景 地震被稱為群災(zāi)之首,其發(fā)生的頻率高、破壞性極強(qiáng),經(jīng)歷地震的人們身心健康可能受到重創(chuàng)。目前強(qiáng)震巨災(zāi)后的心理問題日益引起社會(huì)的廣泛重視,針對(duì)不同的心理問題目前存在不同的干預(yù)手段,音樂療法作為一種新興的治療方式目前在臨床不同科室中得到應(yīng)用,并且已取得了一定的成效。然而音樂療法在強(qiáng)震巨災(zāi)后的心理問題治療方面鮮有報(bào)道。目前科研工作者對(duì)音樂治療的研究主要集中于臨床觀察研究,還缺少詳實(shí)的動(dòng)物實(shí)驗(yàn)和更為先進(jìn)的指標(biāo)。通過動(dòng)物實(shí)驗(yàn)來尋找音樂治療對(duì)疾病病理生理的影響機(jī)制,從而為音樂治療提供一定的實(shí)驗(yàn)室理論依據(jù)。 目的 研究模擬地震應(yīng)激對(duì)大鼠心理行為、血清CORT及腦組織中GR、MR的影響作用,同時(shí)探討早期音樂干預(yù)能否有效改善模擬地震應(yīng)激對(duì)大鼠影響,有望為未來災(zāi)害現(xiàn)場(chǎng)的早期干預(yù)提供理論依據(jù)。 方法 1.應(yīng)用已經(jīng)與北京維爾森科技發(fā)展有限公司成功研制的模擬地震震動(dòng)平臺(tái),可以模擬地震發(fā)生的環(huán)境。10周齡雄性Wistar大鼠30只,體重200g-230g,30只按體重隨機(jī)分為如下三組,C組(control)10只未經(jīng)歷模擬地震刺激的實(shí)驗(yàn)室內(nèi)正常的大鼠；S組(stress):10只大鼠,給予慢性模擬地震刺激30min/d,持續(xù)3w；M組(stress+music):10只大鼠,給予慢性模擬地震刺激30min/d,同時(shí)予音樂治療3h/d,持續(xù)3w。 2.模擬地震刺激結(jié)束后分別于第一天、第二天行曠場(chǎng)行為學(xué)檢測(cè)、懸尾實(shí)驗(yàn)檢測(cè),于第三天活殺各組大鼠,檢測(cè)血清CORT,取腦組織,對(duì)海馬、額葉皮層部位進(jìn)行HE常規(guī)病理切片,并行免疫組織化學(xué)法觀察MR、GR的表達(dá)。 結(jié)果 經(jīng)歷三周模擬地震應(yīng)激后,大鼠血清CORT較正常對(duì)照組明顯升高,差異具有統(tǒng)計(jì)學(xué)意義(P0.05),存在明確的情緒行為改變,主要表現(xiàn)為情緒的焦慮和抑郁(P0.05),海馬、額葉皮層中MR、GR表達(dá)顯著下調(diào)(P0.05)。在給予模擬地震應(yīng)激的同時(shí)進(jìn)行早期音樂干預(yù),大鼠血清CORT較正常對(duì)照組顯著下降(P0.05),MR、GR的表達(dá)明顯上調(diào)(P0.05),而大鼠行為學(xué)的改變未達(dá)到顯著性差異(P0.05)。 結(jié)論 1.慢性模擬地震應(yīng)激可以導(dǎo)致大鼠出現(xiàn)焦慮、抑郁情緒,血清CORT水平升高,海馬、額葉皮層中皮質(zhì)酮受體的表達(dá)下調(diào)。 2.應(yīng)激引起的CORT升高對(duì)海馬、額葉皮層神經(jīng)元具有一定的細(xì)胞毒性作用。 3.早期合理音樂干預(yù)治療可拮抗模擬地震應(yīng)激對(duì)大鼠血清CORT的升高作用,對(duì)大鼠行為的改變有部分改善作用。
[Abstract]:Background earthquakes are called the first mass disasters, their frequency is high and destructive, and the physical and mental health of people who have experienced earthquakes may be severely damaged. At present, the psychological problems after strong earthquake catastrophe have attracted more and more attention of the society. In view of different psychological problems, there are different intervention methods at present. Music therapy, as a new treatment method, has been applied in different clinical departments, and has achieved some results. However, music therapy is rarely reported in the treatment of psychological problems after strong earthquake catastrophe. At present, the research of music therapy is mainly focused on clinical observation and research, and there is still a lack of detailed animal experiments and more advanced indicators. Through animal experiments, we can find out the mechanism of the influence of music therapy on the pathophysiology of diseases, so as to provide a certain laboratory theoretical basis for music therapy.
