2,4-滴丁酯染毒對大鼠學(xué)習(xí)記憶行為及氨基酸類神經(jīng)遞質(zhì)含量的影響
[Abstract]:[aim] to investigate the effects of 2,4-butyl dibutyl on learning and memory behavior and the content of amino acid neurotransmitters in cerebral cortex and hippocampus of male rats. [methods] 40 healthy 30-day-old male Wistar rats were randomly divided into 4 groups: control group (corn oil: distilled water volume ratio was 1:1) and 2,4-dibutyl low (1 渭 100 LD50,6 mg/kg). 2, 4-butyl dibutyl group (3 渭 100 LD50,18 mg/kg), 2,4-butyl dibutyl high dose group (6 渭 100 LD50,36 mg/kg), 10 rats in each group (n = 10, n = 10). Oral administration was given to the stomach for 5 days a week, and the experiment period was 90 d. The learning and memory abilities of rats were tested by Morris water maze at 4,8 and 13 weeks after exposure. After exposure, the contents of glutamic acid, aspartate, 緯-aminobutyric acid and glycine in cerebral cortex and hippocampus of rats were determined by high performance liquid chromatography (HPLC). [results] at the 13th week of Morris water maze test, the mean escape latency of rats gradually prolonged with the increase of dose (P0.05), which showed a dose-effect relationship (r = 0.961 1, P0.01). With the increasing dose of 2,4-dibutyl, the level of glutamic acid in the cerebral cortex increased (P0.05), and the level of 緯-aminobutyric acid decreased (P0.05), both in a dose-effect relationship (r = 0.942 5, P < 0.01, P < 0.01), but the level of 緯-aminobutyric acid (r = 0.942 5, P < 0.05). (r = 0.887 2, P0.01); The content of aspartic acid increased significantly in the high dose group (P0.05). The content of glutamic acid and aspartic acid in hippocampus increased (P0.05), and the content of 緯-aminobutyric acid decreased (P0.05), and all showed a dose-effect relationship (r = 0.9701, r = 0.886 6, r = 0.916 2, P < 0.01), while the content of 緯-aminobutyric acid decreased (P < 0.05). (P0.01). [conclusion] 2, 4-dibutyl subchronic exposure affects the learning and memory ability of rats and the levels of amino acid neurotransmitters in brain tissue of rats.
