DHA干預(yù)對(duì)GDM小鼠胎盤脂肪酸轉(zhuǎn)運(yùn)蛋白基因表達(dá)的影響

【摘要】:目的探討二十二碳六烯酸(DHA)干預(yù)對(duì)妊娠期糖尿病(GDM)小鼠胎盤脂肪酸轉(zhuǎn)運(yùn)蛋白基因表達(dá)的影響。方法選取36只小鼠,其中12只作為空白對(duì)照(CN),24只腹腔注射鏈脲霉素建立GDM模型;GDM小鼠隨機(jī)分為兩組:DHA補(bǔ)充組(GDM+DHA)以及對(duì)照組(GDM)。于第10天~18天,給予GDM+DHA組灌胃DHA 500 mg/kg,其他兩組灌胃等量溶劑,記錄各時(shí)期3組血糖及體重水平,第18天收集孕鼠血清及胎盤。羥胺法和比色法分別測(cè)定母鼠血清超氧化物歧化酶(SOD)和谷胱甘肽過(guò)氧化物酶(GSH-px)活力水平,RT-PCR法測(cè)定胎盤脂肪酸轉(zhuǎn)運(yùn)蛋白基因表達(dá)水平。結(jié)果 (1)重復(fù)測(cè)量資料的方差分析結(jié)果顯示:不同時(shí)間段的孕鼠體重、血糖差異有統(tǒng)計(jì)學(xué)意義(P0.05);不同組別孕鼠血糖差異有統(tǒng)計(jì)學(xué)意義(P0.05)。(2)三組小鼠體內(nèi)SOD活性、GSH-px活力水平差異有統(tǒng)計(jì)學(xué)意義(P0.05);與CN組比,GDM組小鼠體內(nèi)SOD和GSH-px活力水平顯著降低(P0.05),DHA干預(yù)后,小鼠體內(nèi)SOD活力水平顯著升高(P0.05)。(3)與CN組比,GDM下調(diào)了過(guò)氧化物酶體增殖激活受體γ(PPARγ)mRNA表達(dá),上調(diào)脂肪酸結(jié)合蛋白(FABP-pm)、脂肪酸轉(zhuǎn)運(yùn)蛋白-6(FATP-6 mRNA)表達(dá),DHA干預(yù)后,下調(diào)了FABP-pm、FATP-6 mRNA的表達(dá)。結(jié)論DHA干預(yù)可增加GDM小鼠體內(nèi)SOD活力,下調(diào)胎盤FABPpm、FATP-6 mRNA的表達(dá)。
[Abstract]:Objective to investigate the effect of 22 carbohexaenoic acid (DHA) intervention on the expression of fatty acid transporter gene in placenta of (GDM) mice with gestational diabetes mellitus (GDM). Methods 36 mice were selected, 12 of which were used as blank control (CN), 24 mice were injected intraperitoneally with streptozotocin to establish GDM model, GDM mice were randomly divided into two groups: (GDM DHA) supplemented with DHA and (GDM). As control group. From the 10th day to the 18th day, the rats in the GDM DHA group were given DHA 500 mg/kg, and the other two groups were given the same amount of solvent. The blood glucose and body weight of the three groups were recorded. The serum and placenta of the pregnant rats were collected on the 18th day. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-px) in maternal serum were measured by hydroxylamine method and colorimetric method, and the expression level of fatty acid transporter gene in placenta was measured by RT-PCR method. Results (1) the results of ANOVA of repeated measurements showed that the weight of pregnant rats and the difference of blood glucose in different time periods were statistically significant (P0.05). There was significant difference in blood glucose between different groups of pregnant rats (P0.05). (2). There was significant difference in SOD activity and GSH-px activity level among the three groups (P0.05). Compared with CN group, the activity level of SOD and GSH-px in GDM group was significantly lower than that in CN group (P0.05). (3), compared with CN group (P0.05). (3), after intervention with), DHA, the activity level of SOD in mice was significantly higher than that in CN group. GDM down-regulated the expression of peroxisome proliferator-activated receptor 緯 (PPAR 緯) mRNA and up-regulated the expression of fatty acid binding protein (FABP-pm) and fatty acid transporter-6 (FATP-6 mRNA). After DHA intervention, the expression of FABP-pm,FATP-6 mRNA was down-regulated. Conclusion DHA can increase the activity of SOD and down-regulate the expression of placental FABPpm,FATP-6 mRNA in GDM mice.
