砷冶煉廠工人砷甲基化代謝與肝和皮膚損傷及LncRNAs表達(dá)的關(guān)系
[Abstract]:[objective] to investigate the relationship between the metabolic transformation of arsenic methylation and liver and skin injury and the expression of long chain noncoding RNA (LncRNAs) in workers exposed to occupational arsenic. [methods] in October 2013, 112 workers from two arsenic smelters in Wenshan Prefecture, Yunnan Province were selected as exposure group and 41 residents without arsenic exposure as control group. The patients in the contact group were examined for liver and skin injury and were divided into 4 groups (no injury group, liver injury group, skin injury group, liver and skin injury group). The content of inorganic arsenic, methyl arsenic acid and dimethyl arsenic acid in urine was determined by atomic absorption spectrophotometer with arsenic pretreatment system, and the first and second order methylation index was calculated. The expression of LncRNAs (MEG3,TUG1 and HOTAIR) in peripheral blood was detected by real-time fluorescence quantitative PCR. [results] the age of the workers exposed to occupational arsenic was (33.9 鹵16.8) years old and the working age was (16.6 鹵9.7) months. Among them, 42 were in the no-injury group, 29 in the liver injury group, 21 in the skin injury group, and 20 in both the liver and skin injury group. The concentrations of three arsenic compounds in urine of each exposure group were higher than those of the control group (P0.05), and the second order methylation index was lower than that of the control group (P0.05). The first methylation index and the second methylation index of liver injury group and skin injury group were lower than that of liver and skin injury group (P0.05). Compared with the control group, the expression of MEG3 and HOTAIR in peripheral blood was increased in the groups of no injury, skin injury, liver and skin injury, and the expression of TUG1 was increased in the group of liver and skin injury (P0.05). Compared with the liver and skin injury group, the expression of MEG3 was decreased in the non-injury group and the liver injury group, and the expression of TUG1 and HOTAIR was decreased in the liver injury group (P0.05). [conclusion] there is a correlation between the metabolic patterns of arsenic methylation and liver and skin injury, and the expression of LncRNAs in peripheral blood of workers exposed to arsenic.
