十溴聯(lián)苯醚亞慢性染毒對(duì)成年大鼠海馬膽堿能系統(tǒng)和γ-氨基丁酸的影響

【摘要】:目的觀察十溴聯(lián)苯醚(BDE-209)對(duì)成年大鼠海馬膽堿能系統(tǒng)和γ-氨基丁酸(GABA)的影響。方法將64只健康SPF級(jí)SD雄性大鼠隨機(jī)分為4組,溶劑對(duì)照(花生油)組和250、500、1 000 mg/kg BDE-209染毒組,每組16只。采用灌胃方式進(jìn)行染毒,每天1次,連續(xù)染毒60 d。采用Morris水迷宮試驗(yàn)檢測(cè)大鼠的學(xué)習(xí)和記憶能力;采用化學(xué)比色法測(cè)定海馬組織乙酰膽堿轉(zhuǎn)移酶(Ch AT)、乙酰膽堿酯酶(ACh E)的活力和乙酰膽堿(ACh)的含量,采用免疫組化法檢測(cè)海馬組織GABA的表達(dá)情況。結(jié)果與溶劑對(duì)照組比較,在同一訓(xùn)練時(shí)間,各劑量BDE-209染毒組大鼠的逃避潛伏期均延長(zhǎng),跨臺(tái)次數(shù)均減少;海馬組織內(nèi)Ch AT、TCh E活力和ACh含量及CA1區(qū)、CA3區(qū)和DG內(nèi)GABA含量均較低,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。且隨著BDE-209染毒劑量的升高,大鼠的逃避潛伏期呈延長(zhǎng)趨勢(shì),跨臺(tái)次數(shù)呈下降趨勢(shì),海馬組織內(nèi)Ch AT、TCh E活力和ACh含量及CA1區(qū)、CA3區(qū)和DG內(nèi)GABA表達(dá)均呈下降趨勢(shì)。結(jié)論 BDE-209亞慢性染毒導(dǎo)致大鼠學(xué)習(xí)和記憶能力下降,可能與海馬膽堿能系統(tǒng)和GABA表達(dá)下降有關(guān)。
[Abstract]:Objective To observe the effect of ten brominated diphenyl ether (BDE-209) on the hippocampal cholinergic system and gamma aminobutyric acid (GABA) in adult rats. Methods 64 healthy SPF grade SD male rats were randomly divided into 4 groups, solvent control (peanut oil) group and 250500,1 000 mg/kg BDE-209 group, 16 rats in each group, and 1 times a day, 60 d were continuously poisoned by gavage. The Morris water maze test was used to detect the learning and memory ability of rats; the activity of acetylcholine transferase (Ch AT), the activity of acetylcholinesterase (ACh E) and the content of acetylcholine (ACh) in the hippocampus were measured by chemical colorimetry. The expression of GABA in the hippocampus was detected by immunohistochemistry. The results were compared with that of the solvent control group in the same training. The escape latency of the rats in each dose of BDE-209 was prolonged and the number of cross stage decreased, and the content of Ch AT, TCh E activity, ACh content and CA1 area, CA3 and DG GABA content in the hippocampus were lower, and the difference was statistically significant (P0.05). The number of Ch AT, TCh E activity and ACh content and CA1 area in hippocampus, CA3 region and DG in DG are all decreasing. Conclusion BDE-209 subchronic poisoning leads to the decline of learning and memory ability in rats, which may be related to the decrease of hippocampal cholinergic system and GABA expression.
