鱈魚(yú)皮膠原蛋白肽對(duì)小鼠慢性肝損傷的保護(hù)機(jī)制

【摘要】:目的:探究鱈魚(yú)皮膠原蛋白肽對(duì)小鼠慢性肝損傷的保護(hù)作用機(jī)制,為肝病患者功能食品的開(kāi)發(fā)提供理論和試驗(yàn)依據(jù)。方法:采用腹腔注射CCl4建立小鼠慢性肝損傷模型,檢測(cè)鱈魚(yú)皮膠原蛋白肽對(duì)小鼠血清中轉(zhuǎn)氨酶和肝組織勻漿中抗氧化酶以及丙二醛的影響;HE染色觀察小鼠肝組織病理學(xué)變化;蛋白印跡法測(cè)定肝組織Bax,Bcl-2,Cleavage Caspase-3,Caspase-9,Cytosolic Cyt c,TNF-α,JNK,P-JNK蛋白的表達(dá)情況。結(jié)果:與模型組比較,高劑量組(400 mg/kg)小鼠血清中AST、ALT活性、肝組織勻漿中MDA含量顯著降低,而肝組織勻漿中SOD活性顯著升高,且有顯著的劑量關(guān)系;小鼠肝組織損傷情況得到明顯改善;Bcl-2蛋白表達(dá)量增加,Bax,Cleavage Caspase-3,Caspase-9,Cytosolic Cyt c,TNF-α,P-JNK蛋白表達(dá)量降低。結(jié)論:鱈魚(yú)皮膠原蛋白肽可增強(qiáng)慢性肝損傷小鼠的抗氧化活性,抑制氧化應(yīng)激,對(duì)慢性肝損傷有較好的保護(hù)作用。
[Abstract]:Aim: to investigate the protective mechanism of cod skin collagen peptide on chronic liver injury in mice, and to provide theoretical and experimental basis for the development of functional food for patients with liver disease. Methods: the chronic liver injury model of mice was established by intraperitoneal injection of CCl4. The effects of cod collagen peptide on serum aminotransferase, antioxidant enzyme and malondialdehyde in liver tissue homogenate of mice were detected. The pathological changes of liver tissue were observed by HE staining. The expression of Bax-Bcl-2Cleavage Cleavage Caspase-3, cytosolic Cyt and TNF- 偽 in liver tissue was determined by Western blot. Results: compared with the model group, the alt activity in serum and the content of MDA in liver homogenate were significantly decreased in high dose group (400 mg / kg), while the activity of SOD in liver homogenate was significantly increased in a dose-dependent manner. The expression of Bcl-2 protein was significantly improved in mice liver tissue injury. The expression of Cleavage Cleavage Caspase-3, Caspase-9, cytosolic Cyt, TNF- 偽 and P-JNK protein was decreased. Conclusion: Cod skin collagen peptide can enhance the antioxidant activity of chronic liver injury mice, inhibit oxidative stress, and have a better protective effect on chronic liver injury.
