【摘要】:目的研究玻璃酸鈉中間體的遺傳毒性。方法采用鼠傷寒沙門菌/哺乳動物微粒體酶試驗(Ames試驗)、微核試驗、染色體畸變試驗研究玻璃酸鈉中間體的遺傳毒性。結果 (1)Ames試驗:在加與不加大鼠肝微粒體酶(S-9)條件下,玻璃酸鈉中間體在0.8~500μg/皿劑量范圍內(nèi),各菌株回變菌落數(shù)均未超過自發(fā)回變菌落數(shù)的2倍,亦無劑量-反應關系。受試物未引起TA97、TA98、TA100和TA102試驗菌株基因突變,Ames試驗陰性。(2)微核試驗:玻璃酸鈉中間體各劑量組(67、134、268mg/kg)所誘發(fā)的微核率分別為1.7‰、1.7‰和1.9‰,與陰性對照組(1.7‰)比較差異無顯著性,說明在該試驗條件下,該樣品無致小鼠骨髓嗜多染紅細胞微核作用,小鼠骨髓微核試驗呈陰性。(3)中國倉鼠肺細胞染色體畸變試驗:在加與不加S-9兩種條件下,并于24 h后收集細胞進行染色體畸變分析,發(fā)現(xiàn)玻璃酸鈉中間體各劑量組的染色體畸變率與陰性對照組相比差異無顯著性。結論本試驗條件下,玻璃酸鈉中間體Ames試驗結果為陰性,小鼠骨髓嗜多染紅細胞微核試驗結果為陰性,無細胞毒性作用,體外哺乳動物細胞染色體畸變試驗結果為陰性。
[Abstract]:Objective to study the genotoxicity of sodium hyaluronate intermediate.Methods the genotoxicity of sodium hyaluronate intermediate was studied by microsomal enzyme test of Salmonella typhimurium / mammalian microsomal enzyme test, micronucleus test and chromosome aberration test.Results Ames test: under the condition of adding or not increasing the enzyme S-9 of rat liver microsomes, in the dosage range of 0.8 ~ 500 渭 g / dish of sodium hyaluronate, the colony number of each strain was not more than 2 times of the number of spontaneous retrogression colonies, and there was no dose-response relationship.The micronucleus rate induced by sodium hyaluronate intermediate in each dose group was 1.7 鈥,
