【摘要】：目的探討竹蓀醇提物對(duì)LPS誘導(dǎo)的RAW264.7細(xì)胞腫瘤壞死因子(TNF-α)、一氧化氮(NO)的分泌,TNF-α、誘導(dǎo)型一氧化氮合酶(iNOS)、白細(xì)胞介素6(IL-6)、白細(xì)胞介素10(IL-10)的m RNA水平以及核轉(zhuǎn)錄因子(NF-κB)p65蛋白表達(dá)的影響及可能的抗炎機(jī)制。方法以不同濃度(100、200、400μg/ml)的竹蓀醇提物作用于LPS(1μg/ml)誘導(dǎo)的RAW264.7小鼠巨噬細(xì)胞,采用ELISA法和Griess法檢測炎性介質(zhì)TNF-α和NO的分泌量;RT-PCR法測定促炎介質(zhì)TNF-α、iNOS、IL-6和抗炎介質(zhì)IL-10的m RNA水平;Western blot檢測NF-κB p65蛋白的表達(dá)水平。結(jié)果與LPS組相比,200、400μg/ml的竹蓀醇提物能顯著抑制LPS誘導(dǎo)的RAW264.7細(xì)胞TNF-α和NO的釋放(P0.01);且不同程度抑制RAW264.7細(xì)胞中促炎介質(zhì)iNOS、TNF-α和IL-6的m RNA表達(dá)水平(P0.01),劑量依賴性的促進(jìn)抗炎介質(zhì)IL-10的m RNA表達(dá)水平(P0.01);不同濃度的竹蓀醇提物均可顯著抑制LPS誘導(dǎo)RAW264.7細(xì)胞中NF-κB p65蛋白的表達(dá),差異有統(tǒng)計(jì)學(xué)意義(P0.01)。結(jié)論竹蓀醇提物可抑制LPS誘導(dǎo)的RAW264.7細(xì)胞TNF-α和NO的釋放,其抗炎作用的發(fā)揮可能與抑制NF-κB p65蛋白的表達(dá),從而調(diào)節(jié)炎性介質(zhì)的基因水平有關(guān)。
[Abstract]:Objective to investigate the level of m RNA and the transcription factor NF- 魏 B)p65 protein of LPS induced tumor necrosis factor TNF- 偽 (no), inducible nitric oxide synthase (iNOS), interleukin 6 (IL 6) and interleukin 10 (IL 10) in RAW264.7 cells induced by alcohol extract of bamboo sun. Methods LPS(1 渭 g / ml (100 渭 g / ml) was used to induce macrophages in RAW264.7 mice. ELISA and Griess were used to detect the secretion of TNF- 偽 and no. RT-PCR was used to detect the levels of IL-6 and m RNA of TNF- 偽 and IL-10. The expression of NF- 魏 B p65 protein was detected by Western blot. To inhibit the release of TNF- 偽 and no in RAW264.7 cells induced by LPS, and to inhibit the expression level of m RNA of iNOS TNF- 偽 and IL-6 in RAW264.7 cells to varying degrees, and to promote the expression level of m RNA of IL-10 in a dose-dependent manner, and to increase the expression level of m RNA of IL-10 in a dose-dependent manner. The alcohol extract could significantly inhibit the expression of NF- 魏 B p65 protein in RAW264.7 cells induced by LPS. Conclusion the alcohol extract can inhibit the release of TNF- 偽 and no in RAW264.7 cells induced by LPS, and its anti-inflammatory effect may be related to the inhibition of the expression of NF- 魏 B p65 protein and the regulation of the gene level of inflammatory mediators.
