【摘要】：目的:應用卵清蛋白(OVA)建立大鼠變應性鼻炎(AR)模型,探討Toll樣受體(TLR)2和4亞型(TLR2和TLR4)在AR發(fā)病中調(diào)節(jié)非特異性免疫和特異性免疫的作用。方法:80只大鼠隨機分為正常組、AR模型組、AR+脂多糖(LPS)組和AR+肽聚糖(PGN)組,每組20只。HE染色觀察各組大鼠鼻黏膜組織形態(tài)學改變并計數(shù)炎癥細胞浸潤數(shù),免疫組織化學法檢測各組大鼠鼻黏膜組織中TLR2、TLR4和IgE蛋白表達水平,Real-time PCR法檢測各組大鼠鼻黏膜組織中TLR2和TLR4mRNA表達水平。結(jié)果:與正常組比較,模型組、AR+LPS組和AR+PGN組大鼠鼻部癥狀評分和炎癥細胞計數(shù)明顯升高(P0.05)。與正常組比較,模型組大鼠鼻黏膜組織變應性損傷明顯,鼻黏膜以嗜酸性粒細胞浸潤為主,AR+LPS組和AR+PGN組大鼠鼻黏膜組織以中性粒細胞浸潤為主。與正常組比較,模型組、AR+LPS組和AR+PGN組大鼠鼻黏膜組織中TLR2、TLR4和IgE表達水平明顯增高(P0.05);與模型組比較,AR+LPS組大鼠鼻黏膜組織中TLR4和IgE表達水平明顯升高(P0.05);AR+PGN組大鼠鼻粘膜組織中TLR2和IgE的表達水平較模型組也明顯升高(P0.05)。結(jié)論:應用OVA腹腔注射及局部噴鼻能有效建立AR模型,TLR2和TLR4在AR發(fā)病中發(fā)揮調(diào)節(jié)非特異性免疫與特異性免疫的作用。
[Abstract]:AIM: To establish a rat model of allergic rhinitis (AR) by ovalbumin (OVA) and investigate the effects of Toll-like receptor (TLR) 2 and 4 subtypes (TLR2 and TLR4) on regulating non-specific and specific immunity in AR. Methods: 80 rats were randomly divided into normal group, AR model group, AR + lipopolysaccharide (LPS) group and AR + peptidoglycan (PGN) group, with 20 rats in each group. The expression of TLR2, TLR4 and IgE protein in nasal mucosa of rats in each group was detected by immunohistochemistry, and the expression of TLR2 and TLR4 mRNA in nasal mucosa of rats in each group was detected by Real-time PCR. Compared with the normal group, the allergic lesion of the nasal mucosa was obvious in the model group, and eosinophil infiltration was the main infiltration in the nasal mucosa. The infiltration of neutrophils was the main infiltration in the AR + LPS group and the AR + PGN group. The expression levels of TLR2, TLR4 and IgE in nasal mucosa of AR+PGN group were significantly higher than those of model group (P 0.05); the expression levels of TLR4 and IgE in nasal mucosa of AR+LPS group were significantly higher than those of AR+PGN group (P 0.05); the expression levels of TLR2 and IgE in nasal mucosa of AR+PGN group were also significantly higher than those of model group (P 0.05). TLR2 and TLR4 play an important role in regulating nonspecific and specific immunity in AR.
