【摘要】：目的觀察醋酸去氨加壓素(deamino arginine vasopressin,DDAVP)對內淋巴積水模型豚鼠耳蝸水通道蛋白2(aquaporins 2,AQP2)表達的調節(jié),探討梅尼埃病的發(fā)病機制。方法 40只成年豚鼠隨機分為實驗組和對照組,每組各20只。實驗組豚鼠腹腔注射DDAVP 4μg·kg-1·d-1,共7天,構建內淋巴積水模型,對照組豚鼠腹腔注射等量生理鹽水。停藥7天后分別取兩組各5只豚鼠耳蝸切片行HE染色,計算中階與中階加前庭階橫截面積比值平均值(R值)評估內淋巴積水程度,同時,2組各15只豚鼠分別采用免疫組化法和蛋白印跡法檢測耳蝸AQP2蛋白的表達。結果實驗組和對照組的R值分別為0.42±0.04和0.31±0.03,實驗組豚鼠內淋巴積水程度重于對照組(P0.05);實驗組和對照組豚鼠耳蝸AQP2均表達于血管紋、螺旋神經節(jié)細胞,其中,實驗組耳蝸AQP2蛋白半定量分析(0.9954±0.0498)高于對照組(0.7664±0.1452),差異有統(tǒng)計學意義(P0.05)。結論DDAVP上調豚鼠耳蝸中與水轉運密切相關的AQP2的基因表達,參與內淋巴積水形成。
[Abstract]:Objective to investigate the regulation of (deamino arginine vasopressin DDAVP on the expression of aquaporins 2 aquaporin-2 (AQP2) in the cochlea of guinea pigs with endolymphatic hydrops, and to explore the pathogenesis of Meniere's disease. Methods 40 adult guinea pigs were randomly divided into experimental group and control group with 20 in each group. The model of endolymphatic hydrops was established in the experimental group by intraperitoneal injection of DDAVP 4 渭 g kg-1 d -1 for 7 days, and the control group was injected with the same amount of normal saline intraperitoneally. Seven days after withdrawal, the cochlea sections of 5 guinea pigs in each group were stained with HE, and the mean ratio of cross sectional area (R value) of middle and middle order plus vestibular order was calculated to evaluate the degree of endolymphatic hydrops. The expression of AQP2 protein in cochlea was detected by immunohistochemistry and Western blotting in 15 guinea pigs in each group. Results the R values of the experimental group and the control group were 0.42 鹵0.04 and 0.31 鹵0.03, respectively. The degree of endolymphatic hydrops in the experimental group was more serious than that in the control group (P0.05). The cochlear AQP2 was expressed in stria vascularis and spiral ganglion cells in the experimental and control groups. The semi-quantitative analysis of AQP2 protein in cochlea of experimental group (0.9954 鹵0.0498) was higher than that of control group (0.7664 鹵0.1452), the difference was statistically significant (P0.05). Conclusion DDAVP up-regulates the expression of AQP2 related to water transport in guinea pig cochlea and participates in endolymphatic hydrops formation.
