水通道蛋白5與低氧反應(yīng)通路因子(HIF-1α、VEGF)在鼻息肉組織中的表達(dá)及其相關(guān)性研究

【摘要】:目的:檢測(cè)水通道蛋白5(aquaporin-5 , AQP5)、缺氧誘導(dǎo)因子1α( hypoxia-inducible factor-1α, HIF-1α)和血管內(nèi)皮生長(zhǎng)因子( vascular endothelial growth factor, VEGF)在鼻息肉組織中的表達(dá)情況,并探討三種因子之間表達(dá)的相關(guān)性及其在鼻息肉發(fā)病機(jī)制中的作用。 方法:取18例患者鼻息肉組織及10例單純行鼻中隔偏曲矯正術(shù)中切除的下鼻甲組織作為對(duì)照組,分別運(yùn)用Real-time PCR (Real-time Quantitative Polymerase chain Reaction, RT-PCR)和蛋白免疫印跡(Western blot)檢測(cè)AQP5、HIF-1α、VEGF mRNA以及蛋白的表達(dá)情況;采用t檢驗(yàn)和直線相關(guān)分析進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果:①RT-PCR結(jié)果顯示, AQP5mRNA在鼻息肉中的表達(dá)明顯低于下鼻甲組織,差異有統(tǒng)計(jì)學(xué)意義( P 0.01) , HIF-1α、VEGF mRNA在鼻息肉和下鼻甲組織中表達(dá)差異無統(tǒng)計(jì)學(xué)意義( P 0.05) ;②Western blot結(jié)果顯示AQP5蛋白表達(dá)鼻息肉組與對(duì)照組無明顯差異。(P 0.05),HIF-1、VEGF蛋白表達(dá)水平在鼻息肉組織中表達(dá)顯著高于對(duì)照組(P0.05);③Western blot結(jié)果顯示鼻息肉組織中AQP5與HIF-1α之間蛋白的表達(dá)呈明顯正相關(guān)關(guān)系(r =0.633, P 0.01), AQP5與VEGF蛋白質(zhì)表達(dá)亦呈明顯正相關(guān)關(guān)系(r =0.611, P 0.01)。 結(jié)論: AQP5與HIF-1α、VEGF等低氧反應(yīng)通路因子共同參與鼻息肉組織水腫的形成。前者通過組織分布異常引起水轉(zhuǎn)運(yùn)障礙進(jìn)而導(dǎo)致組織水腫形成,后者主要通過誘發(fā)血管增殖、增加血管通透性進(jìn)而導(dǎo)致組織水腫產(chǎn)生;兩者是相對(duì)獨(dú)立的系統(tǒng),我們推測(cè)它們可能通過某種間接途徑發(fā)生聯(lián)系。
[Abstract]:Objective: to investigate the expression of aquaporin-5 (AQP5), hypoxia-inducible factor-1 偽 (HIF-1 偽) and vascular endothelial growth factor (vascular endothelial growth factor,) in nasal polyps. The relationship between the expression of three factors and its role in the pathogenesis of nasal polyps was discussed. Methods: Real-time Polymerase chain Reactionation (RT-PCR) and Western blot (Western blot) were used to detect the expression of AQP5, HIF-1 偽 -VEGF mRNA and protein in 18 cases of nasal polyps and 10 cases of inferior turbinate tissue removed during nasal septum deviation correction. T test and linear correlation analysis were used for statistical analysis. Results the expression of AQP5 mRNA in nasal polyps was significantly lower than that in inferior turbinate tissues (P0.01), but there was no significant difference in HIF-1 偽 -VEGF mRNA expression in nasal polyps and inferior turbinate tissues (P 0.05). 2Western blot showed that the expression of AQP5 protein in nasal polyp group was not significantly different from that in control group. (P 0.05) the expression of HIF-1 protein in nasal polyp tissue was significantly higher than that in control group (P0.05). Western blot showed that AQP5 and HIF-1 偽 in nasal polyp tissue were between AQP5 and HIF-1 偽. The expression of white was positively correlated with that of VEGF (r = 0.633, P 0.01), and the expression of AQP5 was also positively correlated with the expression of VEGF (r = 0.611, P 0.01). Conclusion: AQP5 and HIF-1 偽 -VEGF are involved in the formation of edema in nasal polyps. The former leads to water transport disturbance through abnormal tissue distribution, which leads to the formation of tissue edema, while the latter mainly induces vascular proliferation, increases vascular permeability and leads to tissue edema, which is a relatively independent system. We speculate that they may be connected in some indirect way.
