【摘要】：目的:探討不同修復(fù)條件及修復(fù)液對細胞角蛋白(廣譜)單克隆抗體(Pan)的陽性表達程度的影響,篩選優(yōu)勢抗原修復(fù)方法。方法:對11例鼻腔鼻竇內(nèi)翻性乳頭狀瘤(SNIP)組織石蠟切片,分別采用高溫高壓EDTA修復(fù)、微波EDTA修復(fù)、微波檸檬酸鈉修復(fù)、高溫高壓檸檬酸鈉修復(fù),利用Image-Pro Plus圖像分析儀對每張切片隨機取5個視野,平均計算200個單位染色區(qū)域面積,測定染色區(qū)陽性反應(yīng)的平均吸收度值。結(jié)果:高溫高壓檸檬酸鈉修復(fù)、微波EDTA修復(fù)、微波檸檬酸鈉修復(fù)與高溫高壓EDTA修復(fù)Pan的陽性表達分別為0.324±0.051、0.325±0.056、0.303±0.061及0.365±0.023;前3種方法與高溫高壓EDTA修復(fù)比較,均差異有統(tǒng)計學(xué)意義(均P0.05)。結(jié)論:應(yīng)用高溫高壓EDTA修復(fù)液可最大化增加Pan的陽性表達強度,達到最佳染色結(jié)果,是一種值得推廣的免疫組織化學(xué)修復(fù)方法,可為臨床工作提供更好的指導(dǎo)作用。
[Abstract]:Objective: to investigate the effect of different repair conditions and solutions on the expression of cytokeratin (Broad) monoclonal antibody Pan. Methods: paraffin sections of 11 cases of inverted papilloma of nasal cavity and paranasal sinuses were treated with high temperature and high pressure EDTA repair, microwave EDTA repair, microwave sodium citrate repair, high temperature and high pressure sodium citrate repair, respectively. Image-Pro Plus image analyzer was used to randomly select 5 visual fields for each section. The average area of 200 units of staining area was calculated, and the average absorbance of positive reaction was measured. Results: high temperature and high pressure sodium citrate repair and microwave EDTA repair were used. The positive expression of Pan in microwave sodium citrate repair and high temperature and high pressure EDTA repair were 0.324 鹵0.051 鹵0.325 鹵0.056 鹵0.303 鹵0.061 and 0.365 鹵0.023 respectively. Conclusion: the application of high temperature and high pressure EDTA repair solution can maximize the positive expression intensity of Pan and achieve the best staining result. It is a kind of immunohistochemical repair method worth popularizing. It can provide better guidance for clinical work.
