【摘要】：目的:探討大鼠急性鼻竇炎時嗅黏膜中半胱氨酸天冬氨酸蛋白酶3(Caspase-3)的表達(dá)對嗅神經(jīng)細(xì)胞(olfactory receptor neurons,ORNs)凋亡的影響。方法:將70只成年SD大鼠,按照配對設(shè)計,分成對照組和炎癥組,造模后于不同時間取材,采用免疫組織化學(xué)方法檢測嗅黏膜Caspase-3蛋白表達(dá)情況,原位細(xì)胞DNA斷裂檢測末端標(biāo)記(TUNEL)法觀察ORNs凋亡變化。結(jié)果:免疫組化結(jié)果,對照組ORNs中Caspase-3未見明顯表達(dá),炎癥組ORNs中Caspase-3的表達(dá)在接種細(xì)菌后第2天開始升高,第5天明顯升高(P0.05),第8天達(dá)到高峰(P0.01),第14天逐漸下降,第21天接近正常(P0.05)。細(xì)菌接種后第2、21天,炎癥組TUNEL陽性細(xì)胞計數(shù)與對照組相比沒有統(tǒng)計學(xué)意義(P0.05),第5、8、14天差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論:急性鼻竇炎時Caspase-3的過度表達(dá)可能是促進(jìn)大鼠ORNs凋亡,導(dǎo)致嗅覺功能下降的原因之一。
[Abstract]:Objective: to investigate the effect of the expression of cysteine aspartate protease 3 (Caspase-3) on apoptosis of olfactory receptor neurons1 (ORNs) during acute sinusitis in rats. Methods: 70 adult SD rats were divided into control group and inflammatory group according to paired design. The expression of Caspase-3 protein in olfactory mucosa was detected by immunohistochemical method, and the apoptosis of ORNs was detected by in situ DNA fragmentation and terminal labeling Tunel method. Results: the results of immunohistochemistry were as follows: the expression of Caspase-3 protein in olfactory mucosa was detected by immunohistochemistry, and the expression of ORNs protein in olfactory mucosa was detected by immunohistochemistry. No significant expression of Caspase-3 was found in ORNs of the control group. The expression of Caspase-3 in ORNs of inflammatory group began to increase on the second day after inoculation, increased significantly on the 5th day, reached the peak on the 8th day, decreased gradually on the 14th day, and approached the normal level on the 21st day. The number of TUNEL positive cells in the inflammatory group was not significantly higher than that in the control group (P 0.05), and there was a significant difference between the two groups on the 14th day. Conclusion: the overexpression of Caspase-3 in acute sinusitis may be one of the causes of promoting the apoptosis of ORNs and decreasing the olfactory function in the rats with acute sinusitis.
