【摘要】: 目的通過制備大鼠視網(wǎng)膜光損傷的動物模型,觀察并分析血紅素氧合酶-1(HO-1)對大鼠視網(wǎng)膜光損傷時的細胞保護作用。 方法分別以綠色熒光燈為光源制備光照組模型及光照前30min腹腔注射血晶素(Hemin)制備誘導組模型,以正常大鼠作為對照組,取大鼠視網(wǎng)膜組織,免疫組織化學染色方法觀察HO-1在各組大鼠視網(wǎng)膜組織中的表達情況,以及通過流式細胞術(shù)檢測各組大鼠視網(wǎng)膜細胞的凋亡率。 結(jié)果光照組大鼠在接受光照后較正常對照組大鼠視網(wǎng)膜HO-1的表達有所增加(p0.05),第一天即有表達,第五天時達高峰,隨后逐漸下降,而其視網(wǎng)膜細胞的凋亡率表現(xiàn)出相同的變化趨勢。光照前腹腔注射血晶素(Hemin)的誘導組大鼠視網(wǎng)膜HO-1的表達明顯增高,且各時間點(1d、3d、5d、7d)的表達量均高于光照組(p0.05),而其視網(wǎng)膜細胞的凋亡率較光照組下降,同時間點(1d、3d、5d、7d)凋亡率均低于光照組(p0.05),而高于正常對照組(p0.05)。 結(jié)論血紅素氧合酶-1對光損傷時的視網(wǎng)膜具有明顯的細胞保護作用,且其表達量的水平受其誘導劑的影響而增加。
[Abstract]:Objective to observe and analyze the protective effect of heme oxygenase-1 (HO-1) on retinal light injury in rats. Methods the model of light group was made with green fluorescent lamp and the model of induced group was induced by intraperitoneal injection of hemagglutinin Heminin 30 minutes before illumination. The normal rats were used as the control group and the retinal tissues of the rats were taken out. Immunohistochemical staining was used to observe the expression of HO-1 in the retina of rats in each group and the apoptosis rate of retinal cells in each group was detected by flow cytometry. Results the expression of HO-1 in the retina of the illumination group was higher than that of the normal control group. The expression of HO-1 increased on the first day, reached its peak at 5th days, and then decreased gradually. The apoptosis rate of retinal cells showed the same change trend. The expression of HO-1 in retina of rats induced by intraperitoneal injection of hemagglutinin before illumination was significantly increased. The apoptotic rate of retinal cells was lower than that of light group, and the apoptotic rate of retinal cells was lower than that of light group at the same time. The apoptotic rate of retina cells was lower than that of light group (P 0.05), but higher than that of normal control group (P 0.05). Conclusion heme oxygenase-1 has a significant cytoprotective effect on retina after light injury, and its expression level is increased by its inducer.
