【摘要】:目的觀察形覺剝奪性弱視動物模型應用左旋多巴(levodopa,LD)治療后視皮質17區(qū)生長相關蛋白-43(growth associated protein-43,GAP-43)的表達,探討LD對形覺剝奪性弱視視覺功能恢復作用的可能機制。方法50只4周齡幼貓,隨機分為正常組、剝奪組、對照組、低劑量LD組及高劑量LD組共5組,每組10只。通過單眼眼瞼縫合制備形覺剝奪的弱視模型,觀察不同干預條件下各組圖形視覺誘發(fā)電位(pattern visual evoked potential,PVEP)的P100波的峰時及振幅的變化。應用免疫組織化學法測定各組視皮層17區(qū)GAP-43的差異。結果8周齡時,剝奪組、對照組、低劑量LD組、高劑量LD組剝奪眼P100波振幅比對側眼及同周齡正常組幅值降低,潛時延長(均為P0.05);12周齡時,低劑量LD組剝奪眼P100波振幅增高,與對側眼及同周齡正常組相比差異均無統(tǒng)計學意義(均為P0.05),而P100波潛時則未達到正常(P0.05);高劑量LD組剝奪眼P100波振幅增高、潛時縮短,與對側眼及同周齡正常組相比差異均無統(tǒng)計學意義(均為P0.05)。8周齡時剝奪組視皮質17區(qū)Ⅱ/Ⅲ、Ⅳ、Ⅴ/Ⅵ層GAP-43免疫陽性細胞密度均低于正常組,差異均有統(tǒng)計學意義(均為P0.05);12周齡時高劑量LD組各層GAP-43免疫陽性細胞密度與正常組基本相等,差異無統(tǒng)計學意義(P0.05);對照組、低劑量LD組和正常組相比差異均有統(tǒng)計學意義(均為P0.05)。結論GAP-43在形覺剝奪性弱視形成過程中起一定的作用,LD對形覺剝奪性弱視視功能改善的機制是通過GAP-43表達的改變而實現的。
