【摘要】：目的研究依那普利葉酸對(duì)H型高血壓大鼠外周血炎癥因子水平及頸動(dòng)脈結(jié)構(gòu)的影響。方法按照體重將自發(fā)性高血壓雄性大鼠隨機(jī)分4組:對(duì)照組、模型組、依那普利組以及依葉組,每組各15只。除對(duì)照組外,各組均給予含3%蛋氨酸的飼料喂養(yǎng)建立H型高血壓大鼠模型。模型建立后,依葉組給予10m L·kg~(-1)馬來(lái)酸依那普利葉酸溶液灌胃;依那普利組給予10 m L·kg~(-1)馬來(lái)酸依那普利溶液灌胃;模型組和對(duì)照組分別給予10 m L·kg~(-1)雙蒸水灌胃,均每日1次,連續(xù)給藥8周。用酶聯(lián)免疫法測(cè)定血清同型半胱氨酸(Hcy)、血清腫瘤壞死因子α(TNF-α)、超敏C反應(yīng)蛋白(hs-CRP)及可溶性O(shè)X40配體(OX40L)水平,終點(diǎn)比色法檢測(cè)一氧化氮(NO)水平,顯微鏡下觀察頸動(dòng)脈的中膜厚度(IMT)、頸動(dòng)脈內(nèi)徑(LD)以及頸動(dòng)脈中膜橫截面積(IMCSA)。結(jié)果與依那普利組的血清Hcy與NO水平分別為(64.22±6.82),(41.45±5.25)μmol·L~(-1)相比,依葉組的血清Hcy與NO水平分別為(19.75±2.14),(57.68±6.43)μmol·L~(-1),差異均有統(tǒng)計(jì)學(xué)意義(均P0.05)。與依那普利組的TNF-α、hs-CRP與OX40L分別為(72.72±6.89),(1.15±0.12)pg·m L~(-1)與(5.24±0.47)ng·m L~(-1)相比,依葉組的TNF-α、hs-CRP與OX40L分別為(58.18±5.76),(0.86±0.11)pg·m L~(-1)與(3.03±0.39)ng·m L~(-1),差異均有統(tǒng)計(jì)學(xué)意義(均P0.05)。與依那普利組的頸動(dòng)脈IMT(61.14±7.73)μm、IMCSA水平(0.12±0.02)mm2比較,依葉組大鼠頸動(dòng)脈IMT(52.74±6.29)μm、IMCSA(0.10±0.01)mm2水平,差異均有統(tǒng)計(jì)學(xué)意義(均P0.05)。與依那普利組大鼠頸動(dòng)脈LD水平(951.38±6.08)μm比較,依葉組大鼠頸動(dòng)脈LD水平(969.79±6.17)μm,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論依那普利葉酸能夠明顯降低H型高血壓大鼠外周血Hcy及TNF-α、hs-CRP及OX40L水平,提高NO水平,減輕炎癥反應(yīng),改善頸動(dòng)脈結(jié)構(gòu)。
[Abstract]:Objective to study the effect of enalapril folic acid on peripheral blood inflammatory factor level and carotid artery structure in H-type hypertensive rats. Methods male spontaneously hypertensive rats were randomly divided into 4 groups according to body weight: control group, model group, enalapril group and enalopril group with 15 rats in each group. Except the control group, the H-type hypertensive rat model was established by feeding with 3% methionine. After the establishment of the model, the Enalapril Maleate folic acid solution was given intragastrically in the enalapril group, 10ml kg~ (- 1) enalapril Maleate solution was given intragastrically in the enalapril group, and 10ml kg~ (- 1) double steamed water was given intragastrically once a day for 8 weeks in the model group and the control group, respectively. Serum tumor necrosis factor 偽 (TNF- 偽), high sensitive C reactive protein (hs-CRP) and soluble OX40 ligand (OX40L) were measured by enzyme linked immunosorbent assay (Elisa). Nitric oxide (NO) (no (NO) was measured by end point colorimetric assay. Carotid artery thickness (IMT), carotid artery diameter (LD) and carotid media cross section area (IMCSA). Were observed under microscope. Results compared with enalapril group, serum Hcy and NO levels were (64.22 鹵6.82), (41.45 鹵5.25) 渭 mol 路L ~ (- 1), and serum Hcy and NO levels were (19.75 鹵2.14), (57.68 鹵6.43) 渭 mol 路L ~ (- 1), respectively, and the difference was statistically significant (P 0.05). Compared with enalapril group, TNF- 偽, hs-CRP and OX40L were (72.72 鹵6.89), (1.15 鹵0.12) pg 路mL ~ (- 1) and (5.24 鹵0.47) ng 路mL ~ (- 1), respectively, and TNF- 偽, hs-CRP and OX40L were (58.18 鹵5.76), (0.86 鹵0.11) pg 路mL ~ (- 1) and (3.03 鹵0.39) ng 路mL ~ (- 1), respectively. The difference was statistically significant (P 0.05). Compared with the carotid artery IMT (61.14 鹵7.73) 渭 m and (0.12 鹵0.002) mm2 in the enalapril group, the IMT and mm2 levels in the carotid artery of the enalapril group were (52.74 鹵6.29) 渭 m and (0.10 鹵0.001) 渭 m, respectively (P 0.05). Compared with the level of LD in carotid artery of enalapril group (951.38 鹵6.08) 渭 m, the level of LD in carotid artery of enalapril group was (969.79 鹵6.17) 渭 m, the difference was statistically significant (P 0.05). Conclusion Enalapril folic acid can significantly decrease the levels of Hcy, TNF- 偽, hs-CRP and OX40L in peripheral blood of H type hypertensive rats, increase the level of NO, reduce the inflammatory reaction and improve the structure of carotid artery.
