壓力超負荷大鼠心肌膠原代謝的動態(tài)變化

【摘要】:目的:探討壓力超負荷大鼠心肌膠原代謝的動態(tài)變化。方法:采用腹主動脈縮窄法制備壓力超負荷大鼠模型,并以假手術組為對照,分別于術后第3周、4周、8周和12周取材,稱量大鼠體質量、心臟質量和左心室質量,計算心臟質量指數(shù)(heart mass index,HMI)和左心室質量指數(shù)(left ventricle mass index,LVMI);心肌組織Masson染色觀察心肌膠原纖維沉積情況;樣本堿水解法檢測心肌組織羥脯氨酸(hydroxyproline,HYP)含量;ELISA法檢測血清Ⅰ型前膠原羧基端肽(procollagen typeⅠcarboxy-terminal peptide,PⅠCP)、Ⅲ型前膠原氨基端肽(procollagen typeⅢamino-terminal peptide,PⅢNT)和Ⅰ型膠原C端肽(collagen C telopeptide typeⅠ,CTX-Ⅰ)水平。結果:與假手術組相比,模型組大鼠心肌血管周圍及間質可見膠原纖維沉積,心肌膠原容積分數(shù)明顯增高(P0.01),且隨時間延長膠原沉積明顯增多;模型組大鼠HMI、LVMI和HYP均明顯增加(P0.05),且隨時間延長大鼠HYP有升高的趨勢;模型組大鼠血清PⅠCP濃度顯著升高,在第4、8和12周時差異有統(tǒng)計學意義(P0.05);模型組大鼠血清PⅢNP濃度顯著升高,CTX-Ⅰ濃度顯著降低,差異均有統(tǒng)計學意義(P0.01)。結論:壓力超負荷狀態(tài)下,心肌膠原代謝紊亂,且隨時間的延長心肌呈不同程度的纖維化表現(xiàn)。
