硫化氫對(duì)不同周齡自發(fā)性高血壓大鼠腦基底動(dòng)脈舒張反應(yīng)的影響

【摘要】:目的探討硫化氫(H_2S)對(duì)不同周齡自發(fā)性高血壓大鼠(SHR)離體腦基底動(dòng)脈舒張反應(yīng)的影響。方法選取14、40周齡SHR和正常Wistar京都(WKY)大鼠各6只,測(cè)量血壓后,急性分離腦基底動(dòng)脈,應(yīng)用壓力肌動(dòng)圖技術(shù)檢測(cè)不同濃度硫氫化鈉(NaHS)對(duì)大鼠腦基底動(dòng)脈直徑變化的影響。結(jié)果 (1)10、30、100、300、1 000、3 000 mmol/L的NaHS能夠濃度依賴地舒張SHR和WKY大鼠腦基底動(dòng)脈;(2)與14周齡的WKY相比,1 000、3 000 mmol/L的Na HS對(duì)14周齡SHR的舒張率更明顯,兩者間有顯著性差異(P0.05);(3)與40周齡的WKY大鼠相比,1 000、3 000 mol/L的NaHS對(duì)40周齡SHR的舒張率降低,兩者間有顯著性差異(P0.05)。結(jié)論在高血壓初期,腦基底動(dòng)脈對(duì)H_2S的敏感性增加,而隨著高血壓的發(fā)展及血管功能的嚴(yán)重?fù)p傷,腦基底動(dòng)脈可能對(duì)H_2S的敏感性降低。
[Abstract]:Objective to investigate the effect of hydrogen sulfide (H _ 2S) on diastolic response of isolated cerebral basilar artery in spontaneously hypertensive rats (SHR) of different weeks. Methods six SHR and six normal Wistar Kyoto (WKY) rats aged 14 and 40 weeks were selected. After blood pressure was measured, the cerebral basilar artery was separated. The effect of sodium sulfide (NaHS) at different concentrations on the diameter of cerebral basilar artery in rats was detected by pressure actin technique. Results (1) the basilar arteries of SHR and WKY rats could be dilated in a concentration-dependent manner by NaHS at 10, 30, 100, 000, 000, 000, 000 mmol / L, (2) the relaxation rate of 1 000 ~ 3 000 mmol / L NaHS on SHR at 14 weeks was more obvious than that of WKY at 14 weeks old, and the relaxation rate of NAHS was significantly higher than that of WKY at 14 weeks of age. There was a significant difference between the two groups (P0.05); (3) and 40 weeks old WKY rats compared with the 40 week old WKY rats, the relaxation rate of 10 000 mol / L NaHS was significantly lower than that of 40 week old WKY rats (P0.05). Conclusion in the early stage of hypertension, the sensitivity of cerebral basilar artery to H2S is increased, but with the development of hypertension and the serious injury of vascular function, the sensitivity of cerebral basilar artery to H2S may decrease.
