第一部分CHI3L1在硫代乙酰胺導(dǎo)致的急性肝損傷中的作用目的探討幾丁質(zhì)酶3樣蛋白1（Chitinase-3-like-1,CHI3L1）在硫代乙酰胺（thioacetamide,TAA）導(dǎo)致的急性肝損傷中的作用。方法選擇臨床中存在肝功能損傷的患者（30例）和健康體檢者（10例）,生化儀檢測血清ALT、AST水平,ELISA檢測CHI3L1的水平。利用不同劑量TAA（80mg/kg、150mg/kg）在C57BL/6小鼠體內(nèi)構(gòu)建急性肝損傷模型,3天后利用qRT-PCR檢測肝內(nèi)CHI3L1mRNA的表達(dá),檢測血清ALT、AST、CHI3L1水平。使用均為C57BL/6背景的小鼠建立3組TAA急性肝損傷模型,分別為CHI3L1基因敲除（CHI3L1-KO）組、野生型（WT）組和尾靜脈注射重組CHI3L1蛋白（rCHI3L1）組,檢測三組血清ALT、AST水平,組織病理學(xué)評價(jià)肝臟損傷程度（以Suzuki’s評分量化）。qRT-PCR技術(shù)檢測肝內(nèi)IFN-γ、IL-4、IL-17A和Foxp3的mRNA表達(dá),流式細(xì)胞分析技術(shù)檢測肝內(nèi)CD4⁺IFN-γ⁺、...
