;撬崧(lián)合臍帶間充質(zhì)干細(xì)胞移植修復(fù)酒精性肝損傷

【摘要】:背景:研究表明,臍帶間充質(zhì)干細(xì)胞可以誘導(dǎo)分化成肝樣細(xì)胞,從而對肝損傷有一定的修復(fù)作用。目的:探討;撬崤c人臍帶間充質(zhì)干細(xì)胞移植聯(lián)合治療對肝損傷的修復(fù)作用。方法:48只健康SD大鼠給予56度白酒灌胃30 d建立慢性酒精性肝損傷模型,建模后將其隨機(jī)分為模型組,臍帶間充質(zhì)干細(xì)胞組及;撬+臍帶間充質(zhì)干細(xì)胞組。模型組門靜脈注射生理鹽水1 m L,臍帶間充質(zhì)干細(xì)胞組門靜脈移植1 mL臍帶間充質(zhì)干細(xì)胞懸液,;撬+臍帶間充質(zhì)干細(xì)胞組門靜脈注射牛磺酸0.28 g/L+1 m L臍帶間充質(zhì)干細(xì)胞懸液。移植后2周,檢測各組大鼠血清超氧化物歧化酶、谷胱甘肽過氧化物酶、過氧化氫酶、丙氨酸氨基轉(zhuǎn)移酶以及丙二醛、內(nèi)毒素、白細(xì)胞介素4水平;采用RT-PCR方法檢測肝細(xì)胞特征性功能蛋白CYP2B1的mRNA表達(dá)。結(jié)果與結(jié)論:(1)與模型組相比較,臍帶間充質(zhì)干細(xì)胞組、牛磺酸+臍帶間充質(zhì)干細(xì)胞組超氧化物歧化酶、谷胱甘肽過氧化物酶、過氧化氫酶、谷丙轉(zhuǎn)氨酶水平均顯著增高(P0.05),丙二醛、內(nèi)毒素、白細(xì)胞介素4水平均顯著下降(P0.05)。與臍帶間充質(zhì)干細(xì)胞組比較,;撬+臍帶間充質(zhì)干細(xì)胞組超氧化物歧化酶、谷胱甘肽過氧化物酶、過氧化氫酶、谷丙轉(zhuǎn)氨酶水平進(jìn)一步增高(P0.05),丙二醛、內(nèi)毒素、白細(xì)胞介素4水平進(jìn)一步下降(P0.05);(2)與模型組、臍帶間充質(zhì)干細(xì)胞組比較,肝細(xì)胞特征性功能蛋白CYP2B1的mRNA表達(dá)在;撬+臍帶間充質(zhì)干細(xì)胞組顯著增高,差異有顯著性意義(P0.05);(3)以上實(shí)驗(yàn)結(jié)果表明,臍帶間充質(zhì)干細(xì)胞移植可以修復(fù)大鼠肝損傷,與;撬崧(lián)合使用后效果更佳,其可能與減少自由基對機(jī)體的損害有關(guān)。
[Abstract]:Background: umbilical cord mesenchymal stem cells can induce hepatoid cells and repair liver injury. Objective: to investigate the repair effect of taurine combined with human umbilical cord mesenchymal stem cell transplantation on liver injury. Methods 48 healthy Sprague-Dawley rats were given 56 degrees liquor for 30 days to establish the model of chronic alcoholic liver injury. The model group was randomly divided into three groups: umbilical cord mesenchymal stem cell group and taurine umbilical cord mesenchymal stem cell group. In model group, 1 mL of umbilical cord mesenchymal stem cell suspension was injected into portal vein and 1 mL of umbilical cord mesenchymal stem cell was injected into portal vein of model group and umbilical cord mesenchymal stem cell group. Mesenchymal stem cells were injected with taurine 0.28 g / L -1 mL umbilical cord mesenchymal stem cell suspension via portal vein. The levels of serum superoxide dismutase, glutathione peroxidase, catalase, alanine aminotransferase, malondialdehyde, endotoxin and interleukin-4 were measured 2 weeks after transplantation. The mRNA expression of CYP2B1 was detected by RT-PCR. Results compared with the model group, superoxide dismutase (SOD), glutathione peroxidase (GSH) and catalase were found in the cord mesenchymal stem cell group and taurine cord mesenchymal stem cell group. The levels of glutamate-alanine aminotransferase (alt) increased significantly (P 0.05), and the levels of malondialdehyde (MDA), endotoxin (LPS) and interleukin 4 (IL-4) decreased significantly (P 0.05). Compared with the umbilical cord mesenchymal stem cells group, the levels of superoxide dismutase, glutathione peroxidase, catalase and glutamate-alanine aminotransferase in taurine cord mesenchymal stem cell group were further increased. Compared with model group and umbilical cord mesenchymal stem cell group, the mRNA expression of hepatocyte characteristic protein CYP2B1 was significantly increased in taurine umbilical cord mesenchymal stem cell group. The results showed that the transplantation of umbilical cord mesenchymal stem cells could repair liver injury in rats, and the effect of combined treatment with taurine was better, which might be related to reducing the damage of free radicals to the body.
