【摘要】：目的觀察糖尿病(DM)大鼠結腸動力的變化,并探討其機制。方法將34只SD大鼠隨機分為3組,觀察組和模型組制作DM模型,觀察組于造模6周后每天皮下注射甘精胰島素8 U/kg,對照組和模型組均注射等量生理鹽水。干預4周后處死各組大鼠,剖腹取出全部腸道測量腸道推進率,免疫組化法檢測各組近端結腸Cajal間質細胞(ICC)、縫隙連接蛋白43(Cx43),透射電鏡觀察近端結腸的超微結構。結果觀察組腸道推進率高于模型組,模型組低于對照組,P均0.05。結腸組織ICC及Cx43表達均為對照組高于觀察組、觀察組高于模型組,P均0.05。觀察組ICC數(shù)量、細胞間縫隙連接明顯多于模型組,但少于對照組。結論 DM大鼠存在結腸動力障礙,可能與結腸組織ICC數(shù)量減少、細胞間縫隙連接破壞及Cx43表達降低有關;甘精胰島素可抑制上述改變,改善結腸動力。
[Abstract]:Objective to observe the changes of colon motility in rats with diabetes mellitus (DM) and to explore its mechanism.Methods Thirty-four Sprague-Dawley rats were randomly divided into three groups. DM models were established in the observation group and the model group. The rats in the observation group were subcutaneously injected with insulin glargine (8 U / kg) every day after 6 weeks. The control group and the model group were injected with the same amount of normal saline.After 4 weeks of intervention, all the rats were killed and the intestinal propulsion rate was measured by laparotomy. The interstitial cells of Cajal in the proximal colon of each group were detected by immunohistochemistry, and the ultrastructure of the proximal colon was observed by transmission electron microscope.Results the intestinal propulsion rate of the observation group was higher than that of the model group, and the model group was lower than that of the control group (P < 0.05).The expression of ICC and Cx43 in colon tissue of the control group was higher than that of the observation group, and that of the observation group was higher than that of the model group (P < 0.05).The number of ICC and gap junction between cells in the observation group was significantly higher than that in the model group, but less than that in the control group.Conclusion there is colonic motility disorder in DM rats, which may be related to the decrease in the number of ICC, the destruction of intercellular gap junction and the decrease of Cx43 expression in colon tissue, and insulin glargine can inhibit these changes and improve the colonic motility.
