谷胱甘肽和煙酸對(duì)高糖狀態(tài)下人晶狀體上皮細(xì)胞的影響(英文)

目的:評(píng)價(jià)還原性谷胱甘肽(GSH)和煙酸聯(lián)合應(yīng)用對(duì)蛋白質(zhì)氧化應(yīng)激、內(nèi)質(zhì)網(wǎng)應(yīng)激(ER)、糖基化和聚合αβ晶狀體蛋白的人晶狀體上皮(HLE)細(xì)胞治療高葡萄糖水平的影響。方法:培養(yǎng)HLE細(xì)胞并暴露于25 mmol/L葡萄糖中以促進(jìn)高糖環(huán)境。各組細(xì)胞用三種不同的劑量組合進(jìn)行聯(lián)合治療:10μmol/L GSH+2μmol/L煙酸,30μmol/L GSH+25μmol/L煙酸,100μmol/L GSH+25μmol/L煙酸。培養(yǎng)72h后,用ELISA法測(cè)定蛋白羰基(PCC)和葡萄糖反應(yīng)蛋白(GRP78)的含量。培養(yǎng)2wk后,對(duì)糖基化終末產(chǎn)物(AGEs)進(jìn)行檢測(cè),用Western Blot檢測(cè)αβ晶狀體蛋白的表達(dá)。所有數(shù)據(jù)分析均采用SPSS 18.0統(tǒng)計(jì)軟件包。結(jié)果:與對(duì)照組相比,聯(lián)合治療組的PCC和GRP78水平?jīng)]有顯著降低(P>0.05)。相反,與對(duì)照組相比,GSH和煙酸聯(lián)合治療組的AGEs水平明顯下降(P<0.05)。此外,高劑量葡萄糖給藥后αβ晶狀體蛋白表達(dá)增加,但GSH聯(lián)合治療組與GSH和煙酸聯(lián)合治療組αβ晶狀體蛋白表達(dá)都有所降低。結(jié)論:結(jié)果表明,GSH和煙酸聯(lián)合應(yīng)用可抑制...
