原花青素對(duì)人眼小梁細(xì)胞抗氧化損傷的實(shí)驗(yàn)研究

【摘要】:目的: 研究原花青素(Procyanidins,PC)對(duì)過(guò)氧化氫(Hydrogen peroxide)處理的人眼小梁細(xì)胞(Human Trabecular Meshwork Cells,HTMC)抗氧化損傷的保護(hù)作用,為青光眼的臨床治療提供實(shí)驗(yàn)依據(jù)。 方法: 將正常人眼小梁細(xì)胞進(jìn)行細(xì)胞傳代后隨機(jī)分為5組。未處理組:正常培養(yǎng)的HTMC,對(duì)照組:正常培養(yǎng)的HTMC+過(guò)氧化氫;實(shí)驗(yàn)組:正常培養(yǎng)的HTMC+過(guò)氧化氫+PC(PC濃度分別為0.02mg/ml、0.05mg/ml、0.10mg/ml)。應(yīng)用實(shí)時(shí)熒光定量PCR方法檢測(cè)線粒體復(fù)合物I mRNA(complex Ⅰ mRNA)的表達(dá),流式細(xì)胞術(shù)檢測(cè)細(xì)胞凋亡率,Western blot檢測(cè)細(xì)胞色素C(cytochrome C,CytC)的釋放。 結(jié)果: 1.與對(duì)照組相比,各實(shí)驗(yàn)組線粒體complex Ⅰ mRNA表達(dá)均增加,CytC釋放均減少,細(xì)胞凋亡率均降低,差異均有統(tǒng)計(jì)學(xué)意義(P0.01)。2.不同濃度PC組間隨PC濃度增加complex Ⅰ mRNA表達(dá)增加,CytC釋放減少,細(xì)胞凋亡率降低,各濃度組間差異有統(tǒng)計(jì)學(xué)意義(P0.01)。3.與未處理組相比,各PC組細(xì)胞凋亡率差異均有統(tǒng)計(jì)學(xué)意義(P0.01),,0.02mg/ml和0.05mg/mlPC組complexI mRNA表達(dá)、CytC釋放差異均有統(tǒng)計(jì)學(xué)意義(P0.01),而0.10mg/mlPC組差異均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。 結(jié)論: 外源性PC可以增加氧化損傷人眼小梁細(xì)胞線粒體complex Ⅰ mRNA的表達(dá),減少CytC的釋放,降低細(xì)胞凋亡率,有較強(qiáng)的抗氧化作用,并在一定濃度范圍內(nèi)隨著濃度的增加其抗氧化作用逐漸增強(qiáng),PC在青光眼的臨床治療中的作用值得進(jìn)一步研究。
[Abstract]:Aim: to study the protective effect of proanthocyanidins (Procyanidins,PC) on antioxidant damage of human trabecular meshwork cells (Human Trabecular Meshwork Cells,HTMC) treated with hydrogen peroxide (Hydrogen peroxide) in order to provide experimental basis for clinical treatment of glaucoma. Methods: normal human trabecular meshwork cells were subcultured and randomly divided into 5 groups.
