干擾血管生成素表達(dá)對糖尿病大鼠視網(wǎng)膜視細(xì)胞超微結(jié)構(gòu)的影響

【摘要】:目的探討慢病毒shRNA-血管生成素(ANG)載體干擾ANG表達(dá)對糖尿病大鼠視網(wǎng)膜視細(xì)胞超微結(jié)構(gòu)的影響。方法將40只大鼠隨機(jī)分為對照組、模型組、空載體組及shRNA-ANG組各10只,后三組給予一次性腹腔注射鏈脲佐菌素(STZ)建立糖尿病大鼠模型。造模成功后2周,空載體組及shRNA-ANG組分別向玻璃體腔注射空載體及shRNA-ANG慢病毒載體,其他兩組注射等劑量生理鹽水。造模成功14周時(shí),摘除眼球分離視網(wǎng)膜,檢測視網(wǎng)膜組織ANG mRNA和蛋白相對表達(dá)量,透射電鏡觀察各組視網(wǎng)膜視細(xì)胞超微結(jié)構(gòu)。結(jié)果模型組、空載體組視網(wǎng)膜組織ANG mRNA相對表達(dá)量均高于對照組、shRNA-ANG組(P均0.01),shRNA-ANG組視網(wǎng)膜組織ANG蛋白相對表達(dá)量低于其他三組(P均0.01)。shRNA-ANG組視網(wǎng)膜神經(jīng)細(xì)胞核、線粒體和膜盤超微結(jié)構(gòu)的病變程度均較模型組、空載體組減輕。結(jié)論抑制糖尿病大鼠視網(wǎng)膜ANG表達(dá)可減輕視網(wǎng)膜視細(xì)胞超微結(jié)構(gòu)的病變程度。
[Abstract]:Objective to investigate the effect of (ANG) vector interference of lentivirus shRNA- angiopoietin (shRNA-) on the ultrastructure of retinal visual cells in diabetic rats. Methods Forty rats were randomly divided into control group, model group, empty carrier group and shRNA-ANG group. The latter three groups were given intraperitoneal injection of streptozotocin (STZ) to establish diabetic rat model. Two weeks after the establishment of the model, empty vector and shRNA-ANG lentivirus vector were injected into the vitreous body cavity in the empty vector group and shRNA-ANG group, and the other two groups were injected with the same dose of normal saline. At 14 weeks after the model was successfully made, the eyes were removed to separate the retina, the relative expression of ANG mRNA and protein in the retina tissue was detected, and the ultrastructure of retinal visual cells in each group was observed by transmission electron microscope. Results the relative expression of ANG mRNA in model group and empty carrier group was higher than that in control group and shRNA-ANG group (P0.01). The relative expression of ANG protein in retinal tissue of shRNA-ANG group was lower than that of other three groups (P0.01). The pathological degree of retinal nerve cell nucleus, mitochondria and membrane disc ultrastructure in shRNA-ANG group was less than that in model group and empty carrier group. Conclusion inhibiting the expression of ANG in retina of diabetic rats can reduce the pathological degree of retinal ultrastructure.
