玻璃體腔注射環(huán)孢素A對糖尿病大鼠視網(wǎng)膜功能及結構的保護作用

【摘要】:目的: 從視網(wǎng)膜功能及結構兩方面驗證:早期糖尿病(DM)大鼠即出現(xiàn)視網(wǎng)膜神經(jīng)損害。探討玻璃體腔注射環(huán)孢素A(CsA)對早期DM大鼠視網(wǎng)膜電流圖(ERG)及視網(wǎng)膜組織結構、超微結構的影響。 方法: 健康雄性SD大鼠經(jīng)鏈脲佐菌素誘導建立DM動物模型。 實驗分為正常對照組、未注射藥物組(DM組)、注射CsA干預組(DM+CsA組)、注射對照液干預組(DM+DMSO組),每組9只。 于建模成功后4周進行玻璃體腔注射,注射后48h應用ERG檢測視網(wǎng)膜功能。然后處死大鼠獲取視網(wǎng)膜,制備視網(wǎng)膜HE染色切片及超薄切片,,應用光鏡、透射電鏡檢測視網(wǎng)膜結構變化。 采用GraphPad Prism5統(tǒng)計學軟件進行統(tǒng)計分析。應用采用單因素方差分析ERG結果中a波、b波的振幅及潛伏期數(shù)據(jù)。 結果: (1)與正常對照組相比,3組進行不同干預的DM大鼠均有: ①ERG檢測發(fā)現(xiàn)a波、b波振幅下降; ②視網(wǎng)膜切片HE染色光鏡觀察發(fā)現(xiàn)視網(wǎng)膜外核層疏松變; ③視網(wǎng)膜超薄切片透射電鏡觀察發(fā)現(xiàn)細胞超微結構紊亂呈凋亡表現(xiàn)。 (2)與DM組及DM+DMSO組相比,DM+CsA組表現(xiàn)為: ①ERG a波、b波振幅下降程度輕(P值均<0.05); ②外核層疏松變薄程度輕; ③超微結構改變程度輕。 結論: DM大鼠病程早期即出現(xiàn)視網(wǎng)膜神經(jīng)功能的損害,并有視網(wǎng)膜組織結構紊亂、神經(jīng)細胞超微結構改變。 玻璃體腔內注射CsA可對糖尿病視網(wǎng)膜功能及結構起保護作用。
[Abstract]:Objective: The retinal nerve damage occurred in early diabetic rats. To investigate the effects of intravitreal injection of cyclosporine (CSA) on ERG) and the structure and ultrastructure of retina in early diabetic rats. Methods: DM model was induced by streptozotocin in healthy male SD rats. The experiment was divided into normal control group, non-injection group, CsA intervention group, DM CsA group, and control group, 9 rats in each group. Vitreous injection was performed 4 weeks after successful modeling. ERG was used to detect retinal function 48 hours after injection. Then the rats were sacrificed to obtain the retina and to prepare the HE stained and ultrathin sections of the retina. The structure of the retina was examined by light microscope and transmission electron microscope. GraphPad Prism5 statistical software was used for statistical analysis. The amplitudes and latencies of a wave / b wave in ERG results were analyzed by single factor ANOVA. Results: 1) compared with the normal control group, there were three groups of DM rats with different intervention: The amplitude of a wave B wave was decreased by 1ERG. 2HE staining of retinal sections showed that the outer nuclear layer of the retina became loose and thin; 3 the ultrastructure disorder of the cells showed apoptosis in ultrathin retina sections by transmission electron microscope (TEM). (2) compared with DM group and DM DMSO group, the findings of DM CsA group were as follows: The amplitude of 1ERG a wave and b wave decreased slightly (P < 0.05). 2the degree of loose and thinning of outer nuclear layer is light; 3 the degree of ultrastructure change was slight. Conclusion: The retinal nerve function was damaged in the early stage of diabetic rats, and the retinal tissue structure was disordered and the ultrastructure of nerve cells was changed. Intravitreal injection of CsA can protect the function and structure of diabetic retina.
