Ngn2和Math6在小鼠內(nèi)耳不同時期表達的初步研究

【摘要】:目的研究neurogenin2(ngn2)和math6在不同時期小鼠內(nèi)耳的表達變化,并初步探討其對螺旋神經(jīng)元發(fā)育的調(diào)控作用。方法本實驗選取不同時期健康的BALB/c小鼠,以E12.5、E13.5、E14.5、E17.5、P1、P7和成年為觀察時間點,應(yīng)用real-time PCR、免疫組織化學方法檢測ngn2、math6在耳蝸中mRNA和蛋白的表達。結(jié)果 (1)re al-time PCR結(jié)果提示ngn2在E12.5d呈高表達,隨后在E13.5d表達量下調(diào),直至成年呈低水平表達;math6在E12.5-P1都呈較高水平表達,其中在P1達到高峰,隨后于P7至成年表達量下調(diào)。(2)免疫組織化學結(jié)果提示,在E13.5、E14.5和P1,ngn2和math6在螺旋神經(jīng)節(jié)處(或螺旋神經(jīng)元細胞核處)有不同程度的陽性表達;在成年期,math6在細胞核有弱表達,但ngn2幾乎不表達,各組間差異有統(tǒng)計學意義(P0.05)。結(jié)論 (1)ngn2在E12.5d表達呈高峰,提示其在有增殖能力的神經(jīng)祖細胞中起作用;math6在E12.5d-P1呈高表達,提示其在神經(jīng)祖細胞的增殖以及分化中其起重要作用;(2)ngn2和math6在耳蝸中表達的空間和時間模式的重疊和差異,提示它們在調(diào)控通路中可能受到共同的分子調(diào)控。
[Abstract]:Objective to study the changes of neurogenin 2 ngn2) and math6 expression in the inner ear of mice at different stages, and to explore the regulation of neurogenin 2 ngn2) and math6 on the development of spiral neurons.Methods in this experiment, healthy BALB/c mice in different periods were selected. The expression of mRNA and protein in cochlea was detected by real-time and immunohistochemical method with E12.5E13.5E14.5E14.5E17.5P1P7 and adult as the observation time points. The expression of ngn2mmath6 in the cochlea was detected by immunohistochemical method, and the expression of ngn2mmath6 in the cochlea was detected by immunohistochemistry.Results the results of al-time PCR showed that the expression of ngn2 was high on day 12.5, then down-regulated at day E13.5. until the adult showed a low level of expression of simath6 in E12.5-P1, there was a high level of ngn2 expression in E12.5-P1, and reached its peak at P1.The results of immunohistochemistry showed that there were different degrees of positive expression in the helical ganglion (or the nucleus of the spiral neuron) in E13.5N 14.5, P1ngn2 and math6, and in the nucleus of the spiral neuron in the adult, and in the nucleus of the spiral ganglion, the expression of mafth6 was weakly expressed in the nucleus of adult.However, the expression of ngn2 was almost non-expressed, and the difference was statistically significant (P 0.05).Conclusion the peak expression of ngn2 in E12.5 indicates that it plays an important role in the proliferation of neural progenitor cells and the expression of ngn2 is highly expressed in E12.5d-P1.The results suggest that they play an important role in the proliferation and differentiation of neural progenitor cells. The spatial and temporal patterns of expression of ngn2 and math6 in cochlea may be regulated by common molecules.
