MIF和IL-5在鼻息肉組織中的表達及意義

【摘要】:目的:通過檢測鼻息肉組織中巨噬細胞移動抑制因子(MIF)、白細胞介素-5(IL-5)的表達和嗜酸性粒細胞(Eos)的浸潤情況,并采用視覺模擬量表(VAS)和Lund-Mackay評分法評估鼻息肉患者病情嚴重程度,分析MIF、IL-5表達與Eos浸潤程度間的相關(guān)性及兩因子表達與疾病嚴重程度間的關(guān)系,探討MIF和IL-5在鼻息肉發(fā)生過程中的作用,并分析其可能作用途徑。方法:采用VAS評分和Lund-Mackay評分評估鼻息肉患者病情嚴重程度;應(yīng)用HE染色(蘇木精-伊紅染色法)檢測鼻息肉組織、正常中鼻甲粘膜組織中EOS的浸潤程度并計數(shù);采用免疫組織化學法(SP法)檢測鼻息肉組織、正常中鼻甲組織中MIF、IL-5的表達并進行計數(shù),分析二者與疾病嚴重程度及EOS浸潤程度的相關(guān)性。結(jié)果:1.EOS在鼻息肉組織中的浸潤程度(39.30±4.49)個/HP明顯高于在對照組中的浸潤數(shù)量(7.70±3.19)個/HP,p0.05;2.MIF在鼻息肉組織中的表達(0.3693±0.0535)明顯高于對照組中的表達量(0.1557±0.0395),p0.05;3.IL-5在鼻息肉中的表達量(0.7378±0.2262)明顯高于在對照組中的表達量(0.2253±0.1061),p0.05;4.MIF、IL-5表達與Eos浸潤程度間呈正向相關(guān)性(r分別為0.793、0.843,p均0.05);5.鼻息肉患者VAS評分與MIF、IL-5表達量間呈正相關(guān)性(r分別為0.598、0.769,p均0.05),鼻竇CT Lund-Mackay評分與二者表達同樣呈正相關(guān)性(r分別為0.613、0.626,p均0.05)。結(jié)論:1.MIF、IL-5在鼻息肉組織中的高表達,說明二者共同參與了鼻息肉的發(fā)生發(fā)展;2.MIF、IL-5表達與EOS浸潤程度間呈正相關(guān),說明二者可能通過增加EOS浸潤程度促使鼻息肉的形成;3.MIF、IL-5表達與患者病情嚴重程度間呈正相關(guān),表明二者的表達可在一定程度上評估患者疾病的嚴重程度。
[Abstract]:Objective: to detect the expression of macrophage migration inhibitory factor (MIFN) and interleukin-5 (IL-5) and the infiltration of eosinophil in nasal polyps, and to evaluate the severity of nasal polyps by visual analogue scale (VAS) and Lund-Mackay score. To analyze the relationship between the expression of IL-5 and the degree of Eos invasion, and the relationship between the expression of two factors and the severity of the disease, and to explore the role of MIF and IL-5 in the pathogenesis of nasal polyps. Methods: VAS and Lund-Mackay scores were used to evaluate the severity of nasal polyps, and HE staining (hematoxylin eosin staining) was used to detect nasal polyps. The infiltration degree and count of EOS in normal mucosa of middle turbinate, the expression of MIFIL-5 in nasal polyp tissue and normal middle turbinate tissue were detected and counted by immunohistochemical method. Results 1. The infiltration degree of EOS in nasal polyps was 39.30 鹵4.49) / HP was significantly higher than that in control group (7.70 鹵3.19) and the expression of EOS in nasal polyps was 0.3693 鹵0.0535). The expression of IL-5 in nasal polyps was 0.7378 鹵0.2262), which was significantly higher than that in control group (0.2253 鹵0.1061). The positive correlation between the expression of IL-5 and the degree of Eos infiltration was 0.7930.843 and 0.05 respectively. The positive correlation between the VAS score and the expression of IL-5 in nasal polyps was 0.05% and 0.2253 鹵0.1061 respectively, and the positive correlation between the expression of IL-5 and the degree of Eos infiltration was 0.7930.843, P = 0.05, respectively. The positive correlation between the expression of IL-5 in nasal polyps and the expression of IL-5 in nasal polyps was significantly higher than that in the control group (0.2253 鹵0.1061). There was a positive correlation between 0. 5988 and 0. 769 (p = 0. 05), and there was a positive correlation between CT Lund-Mackay score of paranasal sinus and the expression of both. Conclusion: 1. MIF- IL-5 is highly expressed in nasal polyps, and the positive correlation is 0. 613 ~ 0. 626p, P = 0. 05 respectively. 2. There was a positive correlation between the expression of IL-5 in nasal polyps and the degree of EOS infiltration. It was suggested that the expression of IL-5 in nasal polyps was positively correlated with the severity of the disease by increasing the degree of EOS infiltration. These results suggest that the expression of both can be used to evaluate the severity of the disease to some extent.
