【摘要】：目的研究重樓皂苷Ⅰ對(duì)低氧喉癌Hep-2細(xì)胞增殖及缺氧誘導(dǎo)因子-1α(HIF-1α)、血管內(nèi)皮生長(zhǎng)因子(VEGF)表達(dá)的影響。方法采用MTT法檢測(cè)重樓皂苷Ⅰ對(duì)細(xì)胞增殖的影響,實(shí)時(shí)定量PCR(RT-PCR)法檢測(cè)低氧條件下重樓皂苷Ⅰ對(duì)Hep-2細(xì)胞HIF-1αmRNA、VEGF mRNA表達(dá)的影響。采用Western blot法檢測(cè)低氧條件下,重樓皂苷Ⅰ對(duì)Hep-2細(xì)胞VEGF、HIF-1α、信號(hào)傳導(dǎo)與轉(zhuǎn)錄激活因子3(STAT-3),磷酸化信號(hào)傳導(dǎo)與轉(zhuǎn)錄激活因子3(p-STAT-3)蛋白表達(dá)的影響。結(jié)果重樓皂苷Ⅰ對(duì)低氧喉癌Hep-2細(xì)胞的增殖有抑制作用;其中,1.5、3、6μg/ml濃度組重樓皂苷Ⅰ可下調(diào)低氧Hep-2細(xì)胞HIF-1α、VEGF mRNA和蛋白的表達(dá)。同時(shí),重樓皂苷Ⅰ抑制了STAT-3、p-STAT-3的表達(dá)。結(jié)論重樓皂苷Ⅰ能抑制低氧條件下喉癌Hep-2細(xì)胞增殖和HIF-1α、VEGF的表達(dá),可能與其下調(diào)STAT-3表達(dá)及抑制STAT-3的磷酸化有關(guān)。
[Abstract]:Objective to study the effects of saponins I on the proliferation of Hep-2 cells and the expression of hypoxia inducible factor-1 偽 (HIF-1 偽) and vascular endothelial growth factor (VEGF) in hypoxic laryngeal carcinoma. The effect of saponins I on the expression of HIF-1 偽 mRNA-VEGF mRNA in Hep-2 cells was detected by real-time quantitative PCR- RT-PCR.The Western blot assay was used to detect the expression of HIF-1 偽 -mRNA-VEGF mRNA in Hep-2 cells under hypoxia. The expression of HIF-1 偽, signal transduction and transcriptional activator 3- STAT-3, phosphorylated signal transduction and transcriptional activator 3p-STAT-3) in Hep-2 cells were inhibited. Results the proliferation of Hep-2 cells in hypoxic laryngocarcinoma was inhibited by the expression of SAS-1 偽, phosphorylated signal transduction and transcriptional activator 3p-STAT-3. The expression of HIF-1 偽 -VEGF mRNA and protein in hypoxic Hep-2 cells was down-regulated in 1. 5 渭 g / ml and 6 渭 g / ml groups. At the same time, it inhibited the expression of STAT-3 and p-STAT-3.Conclusion it can inhibit the proliferation of Hep-2 cells and the expression of HIF-1 偽 VEGF in laryngeal carcinoma under hypoxia. It may be related to its down-regulation of STAT-3 expression and inhibition of STAT-3 phosphorylation.
