光交聯(lián)透明質(zhì)酸水凝膠結(jié)合包裹蟲草素殼聚糖治療骨關(guān)節(jié)炎

【摘要】:目的:本研究旨在研究通過使用蟲草素殼聚糖微球(CM-蟲草素)和光交聯(lián)的透明質(zhì)酸甲基丙烯酸酯水凝膠(HAMA)來促進(jìn)軟骨細(xì)胞自噬,評價(jià)此方法治療骨關(guān)節(jié)炎的效果。方法:首先,我們開發(fā)和評價(jià)了 HAMA水凝膠和殼聚糖微球的特性。接下來,我們測量了在由白細(xì)胞介素-1β(IL-1β)誘導(dǎo)的軟骨細(xì)胞和離體模型中蟲草素對軟骨基質(zhì)降解的作用。我們發(fā)現(xiàn)蟲草素可以通過自噬的激活保護(hù)軟骨免于部分降解。此外,我們通過前交叉韌帶切割手術(shù)誘導(dǎo)小鼠骨關(guān)節(jié)炎(OA),并在其關(guān)節(jié)腔內(nèi)注射CM-蟲草素和HAMA。通過組織學(xué)、免疫組織化學(xué)等評價(jià)CM-蟲草素和HAMA水凝膠的組合對手術(shù)誘導(dǎo)的OA的療效。結(jié)果:我們的研究結(jié)果表明:第一,蟲草素能改善離體模型中的軟骨退變;第二,蟲草素能增強(qiáng)小鼠軟骨細(xì)胞和人骨關(guān)節(jié)炎軟骨的自噬;第三,殼聚糖蟲草素和HAMA水凝膠的組合能減緩骨關(guān)節(jié)炎中退行的進(jìn)展。結(jié)論:CM-蟲草素和光交聯(lián)的HAMA水凝膠的組合能明顯改善手術(shù)誘導(dǎo)的骨關(guān)節(jié)炎的進(jìn)展。
[Abstract]:Aim: to study the effect of cordycepin chitosan microspheres (CM- cordycepin) and photocrosslinked hyaluronic acid methacrylate hydrogel (HAMA) on cartilage autophagy and evaluate the effect of this method in the treatment of osteoarthritis. Methods: firstly, we developed and evaluated the properties of HAMA hydrogel and chitosan microspheres. Next, we measured the effect of cordycepin on cartilage matrix degradation in cartilage cells and in vitro models induced by IL-1 尾 (IL-1 尾). We found that cordycepin protects cartilage from partial degradation through autophagy activation. In addition, we induced osteoarthritis (OA), in mice by anterior cruciate ligament cutting and injected CM- cordycepin and HAMA. into their joint cavity. The efficacy of the combination of CM- cordycepin and HAMA hydrogel on surgically induced OA was evaluated by histology and immunohistochemistry. Results: our results showed that: first, cordycepin could improve cartilage degeneration in vitro model, second, cordycepin could enhance autophagy of mouse cartilage cells and human osteoarthritis cartilage. Third, the combination of chitosan cordycepin and HAMA hydrogel can slow down the progress of osteoarthritis. Conclusion: the combination of CM- cordycepin and photocrosslinked HAMA hydrogel can significantly improve the progress of surgery-induced osteoarthritis.
