迷走神經(jīng)電刺激對腦外傷昏迷大鼠意識及前額葉皮質(zhì)γ-氨基丁酸b1受體表達的影響
[Abstract]:Objective to study the effect and mechanism of vagal nerve stimulation (VNS) on brain injury in (TBI) coma rats. Methods 168 healthy Sprague-Dawley rats were randomly divided into control group, TBI group, antagonist group and VNS group with 42 rats in each group. TBI group, VNS group, antagonist group were used to make brain injury model, and the rats in the VNS group were selected to be in coma for at least 30 min. The VNS group was given VNS, antagonist group after injection of Orexin A receptor 1 antagonist SB334867 into the lateral ventricle and VNS, was added to the brain. TBI group was given false VNS. The level of consciousness was observed at 12 h and 24 h after intervention, and the expression of 緯 -aminobutyric acid b1 receptor (GABAb1R) in prefrontal cortex was detected by immunohistochemical, Western blotting. Results 42 rats in the blank group, 11 rats in the TBI group, 13 rats in the antagonist group and 28 rats in the VNS group had awakened. Western blotting. The expression of GABAb1R protein in the control group (F60.412P0.001) was detected 24 hours after the intervention. The expression of GABAb1R protein in the TBI group was significantly higher than that in the control group (F60.412P0.001). Immunohistochemical staining showed that GABAb1R protein was expressed in TBI antagonist group, VNS group was blank group (Hn15.121 P0. 002), but there was no significant difference at different time points (H0. 028, P0. 220). Conclusion VNS can promote the arousal of TBI coma rats, and its mechanism may be related to the down-regulation of GABAb1R expression in prefrontal cortex induced by Orexin A.
