烏頭注射液對兔膝關節(jié)骨性關節(jié)炎關節(jié)液中IL-1β及IL-18含量的影響

【摘要】:目的:通過本課題的實驗研究,探討烏頭注射液對兔膝關節(jié)骨性關節(jié)炎關節(jié)液中IL-1β、IL-18含量的影響,為進一步的臨床應用奠定基礎。方法:1、采選用2%w/v的木瓜蛋白酶溶液與0.03mol/L的L-半胱氨酸溶液比例為2:1的混合溶液,按0.1ml/kg注射至新西蘭大耳白兔雙后膝關節(jié)腔內(nèi),建立兔膝關節(jié)骨性關節(jié)炎模型。2、將20只雄性新西蘭大耳白兔按體重編號隨機分為:空白組、模型組、陽性藥對照組、實驗組。除空白組外,其它組進行造模,2周后行X線觀察造模是否成功。造模成功后,開始治療,實驗組注射烏頭注射液0.2ml,陽性藥對照組注射玻璃酸鈉注射液0.2ml,模型組注射生理鹽水0.2ml,空白組不給予任何處理。治療結(jié)束1周后取雙后膝關節(jié)關節(jié)液,檢測關節(jié)液中IL-1β、IL-18的含量;解剖膝關節(jié)肉眼觀察;經(jīng)HE染色顯微鏡下觀察。3、采用統(tǒng)計學處理分析軟件SPSS19.0對數(shù)據(jù)進行處理,若P0.05即認為檢驗結(jié)果有統(tǒng)計學意義。計數(shù)資料用構(gòu)成比或率的形式表示,需采用x2檢驗;計量資料用x±s形式表示,需采用t檢驗。結(jié)果:1、肉眼觀察軟骨結(jié)果:模型組與空白組比較發(fā)現(xiàn)軟骨表面有多處裂紋或缺損,關節(jié)邊緣幾乎全有骨贅形成,且多呈黃色;陽性藥對照組和實驗組相似,兩者與模型組比較發(fā)現(xiàn)軟骨表面裂紋或缺損較少,少有骨贅形成,且多呈白色。2、顯微鏡下觀察Mankin評分結(jié)果:模型組評分高于空白組(P0.05),差異有統(tǒng)計學意義;陽性藥對照組和實驗組評分均低于模型組(P0.05),差異有統(tǒng)計學意義。3、模型組關節(jié)液中IL-1β、IL-18的含量與空白組比較均明顯增多(P0.05),差異有統(tǒng)計學意義,說明本實驗造模成功;陽性藥對照組和實驗組關節(jié)液中IL-1β、IL-18的含量與模型組比較均明顯降低(P0.05),差異有統(tǒng)計學意義。結(jié)論:烏頭注射液不僅起到保護兔膝關節(jié)軟骨的作用而且能夠降低兔膝關節(jié)骨性關節(jié)炎關節(jié)液中IL-1β、IL-18的含量,達到消除炎癥的目的。
[Abstract]:Objective: to investigate the effect of aconitum injection on the content of IL-1 尾 -IL-18 in the articular fluid of knee joint osteoarthritis of rabbits, so as to lay a foundation for further clinical application. Methods the mixture of 2%w/v 's papain solution and 0.03mol/L 's L-cysteine solution (2:1) was used to inject 0.1ml/kg into the posterior knee joint of New Zealand big ear white rabbit. A rabbit model of osteoarthritis of knee joint was established. Twenty male New Zealand white rabbits were randomly divided into blank group, model group, positive drug control group and experimental group according to their body weight. In addition to the blank group, the other groups were given X-ray after 2 weeks to observe whether the model was successful. After the model was established successfully, the experimental group was injected with aconitum injection 0.2 ml, the positive control group with sodium hyaluronate injection 0.2 ml, the model group with physiological saline 0.2 ml, the blank group without any treatment. After 1 week of treatment, the contents of IL-1 尾 -IL-18 in the joint fluid were measured, the naked eyes of the knee joint were dissected, observed under the microscope of HE staining, and the data were processed by the statistical analysis software SPSS19.0. If P0.05 that the test results were statistically significant. The counting data is expressed in the form of composition ratio or rate, which needs x2 test, and the measurement data is expressed in the form of x 鹵s, and the t test is used. Results: compared with the blank group, the model group found that there were many cracks or defects on the cartilage surface, almost all the osteophytes formed on the articular edge, and most of them were yellow, while the positive drug control group was similar to the experimental group. Compared with the model group, it was found that the cartilage surface cracks or defects were less, few osteophytes formed, and most of them were white. The Mankin score of the model group was higher than that of the blank group (P0.05), and the difference was statistically significant. The score of positive drug control group and experimental group were lower than that of model group (P0.05), the difference was statistically significant. The content of IL-1 尾 -IL-18 in joint fluid of model group was significantly higher than that of blank group (P0.05), the difference was statistically significant. The content of IL-1 尾 -IL-18 in the joint fluid of the positive control group and experimental group was significantly lower than that of the model group (P0.05), and the difference was statistically significant. Conclusion: aconitum injection can not only protect the articular cartilage of rabbit knee joint, but also reduce the content of IL-1 尾 -IL-18 in the articular fluid of rabbit knee osteoarthritis, so as to eliminate inflammation.
