姜黃素對大鼠退行性腰椎間盤NF-κB表達的影響

【摘要】:目的觀察姜黃素對大鼠退行性腰椎間盤髓核中核轉錄因子-kappa B-p65(NF-κB-p65)、腫瘤壞死因子-alpha(TNF-a)表達的影響并且探討其延緩椎間盤退行性改變進程機制的研究。方法建立Sprague-Dawley(SD)大鼠腰椎間盤退行性改變模型,給予姜黃素治療,超顯微組織形態(tài)學變化評價SD大鼠腰椎間盤退行性改變,分別檢測退變的椎間盤組織中NF-κB-p65與TNF-a在蛋白和基因的表達。結果1.通過腰椎核磁共振(MRI)和組織形態(tài)學證實大鼠腰椎間盤退變模型建立成功;2.通過MRI和超顯微結果發(fā)現(xiàn)姜黃素治療組可以明顯延緩腰椎間盤退變(低劑量和高劑量),在溶劑對照組(dimethyil sulfoxide,DMSO)中觀察并沒有顯著改變;3.根據(jù)Western blot和real time PCR結果分析,姜黃素治療組(低劑量和高劑量)與未給予姜黃素治療組相比,前者能夠減少退變腰椎間盤組織中的NF-κB-p65、TNF-a的表達;4.電鏡中觀察、Western blot和real time PCR結果分析,姜黃素低劑量組和高劑量組之間沒有差異。結論高劑量的姜黃素可有效抑制腰椎間盤的退變程度、減少膠原纖維的損傷,并可抑制退變的椎間盤中NF-ΚB-p65活性、降低TNF-alpha的表達水平。
[Abstract]:Objective To observe the effect of curcumin on the expression of nuclear factor -kappa B-p65 (NF- kappa B-p65) and tumor necrosis factor -alpha (TNF-a) in the degenerative nucleus pulposus of rats and to explore the mechanism of delaying the process of degeneration of intervertebral disc. Methods to establish the degenerative change model of the lumbar intervertebral disc in Sprague-Dawley (SD) rats and give curcumin. Treatment, ultrastructural changes of ultrastructure to evaluate the degenerative changes of lumbar intervertebral disc in SD rats and the expression of NF- kappa B-p65 and TNF-a in the degenerative intervertebral disc respectively. Results 1. the lumbar disc degeneration model was successfully established by lumbar nuclear magnetic resonance (MRI) and histomorphology, and 2. through MRI and ultramicro results. It was found that the curcumin treatment group could significantly delay the lumbar intervertebral disc degeneration (low dose and Gao Jiliang), and no significant changes were observed in the solvent control group (dimethyil sulfoxide, DMSO); 3. according to the results of Western blot and real time PCR, the curcumin treatment group (low dose and high dose) could be compared with the non curcumin treatment group. Reduction of NF- kappa B-p65, TNF-a expression in degenerative lumbar intervertebral disc; 4. electrical mirror observation, Western blot and real time PCR results, no difference between curcumin low dose group and high dose group. Conclusion high dose curcumin can effectively inhibit the degree of degeneration of lumbar intervertebral disc, reduce the damage of collagen fiber, and inhibit the degenerative vertebra. The activity of NF- B-p65 in the pan decreased the expression level of TNF-alpha.
