【摘要】：目的 探究環(huán)氧合酶(Cox)-2選擇性抑制劑塞來昔布對(duì)大鼠創(chuàng)傷性異位骨化的防治機(jī)制。方法 創(chuàng)傷性異位骨化大鼠予塞來昔布治療,分別采取喂食2、6、10 w的不同方案,治療后觀察各組肢體的皮溫、腫脹程度及X線表現(xiàn),并通過實(shí)時(shí)定量PCR及Western印跡實(shí)驗(yàn),檢測(cè)跟腱組織骨形態(tài)發(fā)生蛋白(BMP)-4的表達(dá)含量。結(jié)果 塞來昔布組皮溫、腫脹程度、新骨形成均顯著低于對(duì)照(生理鹽水處理)組(P0.05)。同時(shí),隨著塞來昔布處理時(shí)間的延長(zhǎng),皮溫均明顯降低、腫脹程度及新骨形成情況均明顯減輕;實(shí)時(shí)定量PCR及Western印跡檢測(cè)顯示:與對(duì)照組比較,塞來昔布組跟腱組織BMP-4的表達(dá)明顯下調(diào),并且隨時(shí)間的延長(zhǎng),BMP-4的表達(dá)也相應(yīng)下調(diào)。結(jié)論 塞來昔布可通過降低大鼠跟腱組織BMP-4的表達(dá)以干預(yù)異位骨化。
[Abstract]:Objective to investigate the protective mechanism of celecoxib, a selective inhibitor of cyclooxygenase-2, on traumatic ectopic ossification in rats. Methods Rats with traumatic ectopic ossification were treated with celecoxib for 10 weeks. The skin temperature, swelling degree and X ray appearance of each group were observed, and real-time quantitative PCR and Western blot were used. The expression of bone morphogenetic protein (BMP- 4) in Achilles tendon was detected. Results the skin temperature, swelling and new bone formation in celecoxib group were significantly lower than those in control group (P 0.05). At the same time, with the prolongation of celecoxib treatment time, the skin temperature was significantly decreased, the swelling degree and new bone formation were significantly reduced, and real-time quantitative PCR and Western blot analysis showed that: compared with the control group, In celecoxib group, the expression of BMP-4 in Achilles tendon was down-regulated, and the expression of BMP-4 was down-regulated with time. Conclusion Celecoxib can interfere with ectopic ossification by decreasing the expression of BMP-4 in Achilles tendon.
