【摘要】：目的探討趨化因子CXCL13對(duì)TNF-α誘導(dǎo)的人骨關(guān)節(jié)滑膜細(xì)胞調(diào)控細(xì)胞核因子-κB受體活化因子配體(RANKL)表達(dá)的影響。方法 (1)體外培養(yǎng)人骨關(guān)節(jié)滑膜細(xì)胞,并給予10μg/m L TNF-α處理,分別在培養(yǎng)0、6、12、24 h時(shí)采用免疫熒光法檢測(cè)CXCL13表達(dá)。(2)人滑膜細(xì)胞給予10μg/m L TNF-α處理24 h,將培養(yǎng)液更換為含0、5、10、25 ng/m L CXCL13的培養(yǎng)液繼續(xù)培養(yǎng)24 h,或?qū)⑴囵B(yǎng)液更換為含25 ng/m L CXCL13的培養(yǎng)液分別作用0、1、3、6、24 h;以不加TNF-α及CXCL13處理的常規(guī)培養(yǎng)細(xì)胞作為對(duì)照細(xì)胞。收集各濃度、各時(shí)間點(diǎn)細(xì)胞,采用Western blotting法檢測(cè)RANKL蛋白相對(duì)表達(dá)量。結(jié)果 (1)TNF-α作用0、6、12、24 h時(shí)細(xì)胞CXCL13相對(duì)表達(dá)量(熒光強(qiáng)度)分別為0.907±0.350、0.823±0.730、0.710±0.660、0.653±0.850,組間比較P均0.05。(2)對(duì)照細(xì)胞RANKL蛋白相對(duì)表達(dá)量為0.956±0.014,25 ng/m L CXCL13處理0、1、3、6、24 h時(shí)RANKL蛋白相對(duì)表達(dá)量分別為1.543±0.047、1.366±0.026、0.883±0.026、0.367±0.034、0.246±0.015;0、5、10、25 ng/m L CXCL13作用24 h時(shí)RANKL蛋白相對(duì)表達(dá)量分別為0.287±0.007、0.189±0.008、0.069±0.004、0.022±0.002;上述各組、各時(shí)間及各濃度細(xì)胞比較P均0.05。結(jié)論趨化因子CXCL13能夠在一定濃度和時(shí)間內(nèi)抑制TNF-α誘導(dǎo)的人骨關(guān)節(jié)滑膜細(xì)胞RANKL蛋白表達(dá)。
[Abstract]:Objective to investigate the effect of chemokine CXCL13 on the expression of nuclear factor- 魏 B receptor activating factor ligand RANKL in human osteoarticular synovial cells induced by TNF- 偽. Methods Human osteoarticular synovial cells were cultured in vitro and treated with 10 渭 g / mL TNF- 偽.
