低氧環(huán)境下丙泊酚可增加PC12細(xì)胞的凋亡

【摘要】:目的探討低氧環(huán)境下丙泊酚對PC12細(xì)胞株凋亡的影響及機(jī)制。方法將PC12細(xì)胞接種于培養(yǎng)板中,采用隨機(jī)數(shù)字表法,將其隨機(jī)分為6組:低氧對照組(CH組)、空氣對照組(CA組)和氧氣對照組(CO組),對照組加入脂肪乳濃度10μmol/L。丙泊酚低氧組(PH組)、丙泊酚空氣組(PA組)和丙泊酚氧氣組(PO組),丙泊酚濃度為10μmol/L。藥物處理完畢后分別放入低氧(5%O_2),空氣和氧氣(35%O_2)環(huán)境的細(xì)胞培養(yǎng)箱培養(yǎng),8 h后流式檢測細(xì)胞凋亡情況,檢測細(xì)胞內(nèi)ROS水平和細(xì)胞SOD酶活力。結(jié)果與CA組比較,PA組細(xì)胞凋亡增加,活性氧簇(ROS)升高,超氧化物歧化酶(SOD)活力增強(qiáng);與CH組比較,PH組細(xì)胞凋亡增加,ROS升高,SOD活力增強(qiáng);與CO組比較,PO組細(xì)胞凋亡增加,ROS升高,SOD活力增強(qiáng);與PA組比較,PH細(xì)胞凋亡增加,ROS升高,SOD活力增強(qiáng);CO組、CA組和CH組上述各指標(biāo)差異無統(tǒng)計學(xué)意義。結(jié)論低氧環(huán)境下丙泊酚可通過氧化應(yīng)激損傷導(dǎo)致PC12細(xì)胞凋亡增加。
[Abstract]:Objective to investigate the effect and mechanism of propofol on apoptosis of PC12 cell line under hypoxia. Methods PC12 cells were inoculated on the culture plate and randomly divided into 6 groups: hypoxia control group (Ch group), air control group (CA group) and oxygen control group (CO group). The control group was supplemented with 10 渭 mol / L fat milk. The concentration of propofol was 10 渭 mol / L in propofol hypoxia group, propofol air group (PA group) and propofol oxygen group (10 渭 mol / L). After the drug treatment, the cells were cultured in a cell incubator with hypoxia 5 and O _ 2, air and oxygen, respectively. After 8 hours of culture, the apoptosis of cells was detected by flow cytometry, and the level of ROS and the activity of SOD enzyme in the cells were detected. Results compared with CA group, apoptosis increased, Ros increased, SOD activity increased in PA group and SOD activity in PH group. Compared with CO group, the apoptosis increased in PO group and the activity of SOD was increased in Ros group, but there was no significant difference in the above indexes between CA group and Ch group in CO group compared with PA group. Conclusion propofol can induce apoptosis of PC12 cells through oxidative stress injury.
