六磷酸肌醇對人骨肉瘤細胞株MG-63和原代培養(yǎng)成骨細胞增殖的影響

【摘要】:目的研究六磷酸肌醇(inositol hexaphosphate,IP6)對人骨肉瘤細胞株MG-63和原代培養(yǎng)成骨細胞增殖的影響,并探求其抗癌效果的最佳作用濃度。方法原代培養(yǎng)人成骨細胞并鑒定,復蘇并常規(guī)培養(yǎng)人骨肉瘤細胞株MG-63,MTT法檢測不同濃度IP6干預培養(yǎng)時兩種細胞的增殖情況,確定IP6的最佳抑癌濃度,并進一步用流式細胞儀檢測MG-63細胞周期及凋亡情況。結(jié)果當IP6濃度≥1mmol/L時開始抑制MG-63細胞增殖,在4mmol/L時達到最大,并呈時間-劑量依賴性;當IP6濃度為0.5、1mmol/L時,對成骨細胞增殖抑制不明顯,2mmol/L時輕度抑制,4mmol/L時導致成骨細胞凋亡。結(jié)論 IP6能夠抑制人骨肉瘤細胞MG-63的增殖并導致其凋亡,其抗癌的最佳作用濃度為2mmol/L。
[Abstract]:Objective to study the effects of inositol hexaphosphate IP6) on the proliferation of human osteosarcoma cell line MG-63 and primary cultured osteoblasts, and to find out the best concentration of its anticancer effect.Methods Primary culture of human osteoblasts and identification, resuscitation and routine culture of human osteosarcoma cell line MG-63 were used to detect the proliferation of human osteosarcoma cells treated with different concentrations of IP6, and to determine the best inhibitory concentration of IP6.Furthermore, MG-63 cell cycle and apoptosis were detected by flow cytometry.Results when the concentration of IP6 鈮,
