BMP誘導(dǎo)活體內(nèi)肌腱組織成骨化的研究

【摘要】:[目的]探討活體內(nèi)BMP誘導(dǎo)肌腱組織成骨化的效果。[方法]30只新西蘭兔建立活體內(nèi)肌腱組織誘導(dǎo)成骨模型,一側(cè)為rh BMP-2處理、對(duì)側(cè)為空白對(duì)照。術(shù)后4、8及12周時(shí)采用X線(xiàn)片檢測(cè)肌腱組織鈣化情況,術(shù)后12周時(shí)對(duì)肌腱組織進(jìn)行大體解剖、組織形態(tài)及免疫組化檢測(cè)分析。[結(jié)果]術(shù)后4、8及12周時(shí)X線(xiàn)片檢測(cè)發(fā)現(xiàn)BMP處理側(cè)各時(shí)間點(diǎn)上肌腱組織內(nèi)可見(jiàn)鈣化區(qū)域,成骨化范圍隨時(shí)間增加逐漸增大(5.51 mm2、9.44 mm2及20.26mm2,P0.05),鈣化區(qū)域IOD值亦隨時(shí)間增加而增高(1 178.56、2 135.26及4 726.72,P0.05)。大體解剖可見(jiàn)BMP處理組肌腱組織增粗、質(zhì)地變硬,H.E及Masson染色檢測(cè)見(jiàn)肌腱纖維內(nèi)細(xì)胞浸潤(rùn),局部區(qū)域骨樣組織形成,并有osteonectin陽(yáng)性表達(dá)細(xì)胞分布。此外,肌腱纖維及骨化區(qū)域內(nèi)均可見(jiàn)軟骨樣細(xì)胞形成。[結(jié)論]BMP能夠誘導(dǎo)活體內(nèi)肌腱組織發(fā)生成骨化,可能與軟骨內(nèi)成骨途徑有關(guān)。
[Abstract]:[objective] to investigate the effect of BMP on ossification of tendon tissue in vivo.[methods] 30 New Zealand rabbits were used to establish osteogenic model of tendon tissue in vivo, one side was treated with rh BMP-2, the other side was blank control.The calcification of tendon tissue was detected by X-ray film at 8 and 12 weeks after operation, and the gross anatomy, histological morphology and immunohistochemical analysis were made at 12 weeks after operation.[results] X-ray examination at 8 and 12 weeks after BMP showed that calcification areas could be seen in the superior tendons of the side treated with BMP at different time points.The range of ossification gradually increased with time, 5.51 mm ~ 2 ~ 9.44 mm2 and 20.26 mm ~ (2) ~ (2) P _ (0.05), and the IOD value of calcified area increased with time. The values of IOD in calcified area increased by 11178.56 ~ (2) 135.26 and 4 726.72 ~ (2) (P _ (0.05)).Gross anatomy showed that the tendon tissue in BMP group was thicker, the texture of tendon was stiffened, and the infiltration of cells in tendon fiber and the formation of bone like tissue in local area were detected by using H. E and Masson staining. The positive expression of osteonectin was also found in the distribution of osteonectin positive cells.In addition, chondroid cells were found in tendon fibers and ossification areas.[conclusion] BMP can induce ossification of tendon tissue in vivo, which may be related to endochondral osteogenesis.
