大鼠脊髓壓迫性損傷解壓后pEGFR的表達(dá)變化

【摘要】:目的探討大鼠脊髓壓迫性損傷(compressed spinal cord injury,CSCI)解壓后表皮生長因子受體(phosphorylated epidermal growth factor receptor,pEGFR)、pAkt1的表達(dá)變化及其與神經(jīng)功能、有髓神經(jīng)纖維數(shù)量變化的相關(guān)性,為CSCI解壓后治療策略的制定和藥物的研發(fā)提供實(shí)驗(yàn)基礎(chǔ)。方法采用自行設(shè)計(jì)的方法制作SD大鼠CSCI模型,造模成功后解壓。運(yùn)用BBB(Basso Beattie Bresnahan)評(píng)分觀察動(dòng)物解壓后神經(jīng)功能的恢復(fù)情況;通過Luxol fast blue(LFB)染色檢測(cè)解壓后1、7、14、21 d有髓神經(jīng)纖維數(shù)量變化;運(yùn)用免疫熒光雙標(biāo)(Double-labeling immunoflurescence)、免疫印跡(western blotting,WB)檢測(cè)pEGFR、pAkt1的表達(dá)變化。結(jié)果 CSCI解壓后,BBB評(píng)分和有髓神經(jīng)纖維數(shù)量隨時(shí)間延長而逐漸增加;與此同時(shí),pEGFR、pAkt1表達(dá)亦上調(diào)且與BBB評(píng)分、有髓神經(jīng)纖維數(shù)量增加趨勢(shì)一致。結(jié)論CSCI解壓后,神經(jīng)功能有一定改善、有髓神經(jīng)纖維數(shù)量有一定增加,這些變化與pEGFR表達(dá)上調(diào)有關(guān),提示EGFR的活化參與了CSCI解壓后的內(nèi)源性修復(fù)。
[Abstract]:Objective to investigate the changes of EGF receptor phosphorylated epidermal growth factor receptor pEGFRR pAkt1 after decompression of compressed spinal cord injury-induced spinal cord injury in rats and its correlation with the changes of nerve function and the quantity of myelinated nerve fibers. Methods SD rat CSCI model was made by using self-designed method, and the model was successfully established. The recovery of nerve function after decompression was observed by BBB(Basso Beattie Bresnahan score. The number of myelinated nerve fibers was detected by Luxol fast blue LFB staining on the 1st day, 1421 d after decompression. The expression of pEGFRN pAkt1 was detected by double labeling and Western blotting with immunofluorescence. Results the CSCI score and the number of myelinated nerve fibers increased gradually with time after decompression, and the expression of pEGFRR pAkt1 was also up-regulated and correlated with BBB score. Conclusion the number of myelinated nerve fibers and the number of myelinated nerve fibers increased after CSCI decompression. These changes are related to the upregulation of pEGFR expression. The results suggest that the activation of EGFR is involved in the endogenous repair after decompression of CSCI.
