【摘要】：目的:探索淫羊藿苷對大鼠肩袖損傷重建術(shù)后腱骨愈合的影響。方法:健康雄性Sprague Dawley(SD)大鼠(體重250-300g)64只,隨機(jī)分為淫羊藿苷組(即ICA組,n=32)和對照組(n=32)。將岡上肌肌腱于肱骨大結(jié)節(jié)止點(diǎn)處切斷后采用骨隧道法原位縫合,建立大鼠肩袖損傷重建模型。ICA組大鼠術(shù)后每天以125mg/kg淫羊藿苷灌胃,對照組中大鼠每日給予相同劑量的生理鹽水灌胃。兩組分別于術(shù)后2周、4周各處死一半大鼠(每組16只)并收集標(biāo)本,通過Micro-CT掃描、血清生化、生物力學(xué)和組織學(xué)檢測等手段評估淫羊藿苷對大鼠肩袖損傷重建術(shù)后腱骨愈合的影響。結(jié)果:血清生化檢測提示ICA組術(shù)后2周、4周堿性磷酸酶及鈣蛋白濃度顯著高于對照組(P0.05)。Micro-CT掃描提示ICA組骨密度及骨體積分?jǐn)?shù)(BV/TV)顯著高于對照組(P0.05)。與對照組相比,ICA組腱骨交界檢測到更多的軟骨樣細(xì)胞、成骨細(xì)胞、Collagen Ⅰ和VEGF陽性細(xì)胞。生物力學(xué)測定發(fā)現(xiàn)ICA組最大抗拉強(qiáng)度大于對照組(P0.05)。兩組標(biāo)本術(shù)后各時(shí)間點(diǎn)Collagen Ⅲ的表達(dá)水平無顯著差異(P0.05)。結(jié)論:實(shí)驗(yàn)結(jié)果提示淫羊藿苷可促進(jìn)腱骨交界區(qū)域血管及新骨形成并可增強(qiáng)肩袖重建術(shù)后力學(xué)性能,對大鼠肩袖損傷重建術(shù)后腱骨早期愈合有潛在促進(jìn)作用。
[Abstract]:Objective: to investigate the effect of icariin on tendon healing after rotator cuff reconstruction in rats. Methods: healthy male Sprague Dawley SD rats (body weight 250-300g)64) were used in this study. Icariin group (ICA group) and control group (control group) were randomly divided into two groups. The supraspinatus muscle tendon was severed at the insertion point of the great tubercle of humerus and then sutured in situ with bone tunnel method to establish the rotator cuff injury reconstruction model of rats. ICA group rats were perfused with 125mg/kg icariin every day after operation. Rats in the control group were given the same dose of normal saline daily. Half of the rats (16 rats in each group) died 2 weeks and 4 weeks after operation, and the samples were collected, and the serum biochemistry was detected by Micro-CT scanning. Effects of icariin on tendon healing after rotator cuff reconstruction in rats were evaluated by biomechanical and histological methods. Results: serum biochemical examination showed that alkaline phosphatase and troponin concentrations in ICA group were significant at 2 weeks and 4 weeks after operation. Compared with control group, bone mineral density and bone volume fraction of ICA group were significantly higher than that of control group (P 0.05). More chondroid cells were detected in tendon bone junction of ICA group than those in control group. The maximum tensile strength of ICA group was higher than that of control group (P0.05).
