【摘要】：目的探討經(jīng)尾靜脈注射大鼠骨髓間充質(zhì)干細(xì)胞(BMMSC)對(duì)不同品系小鼠移植皮膚存活時(shí)間的影響及機(jī)制。方法分離SD大鼠BMMSC培養(yǎng)至第3代。用皮膚取樣器切取60只ICR乳鼠圓形皮片(直徑12 mm),將其移植至成年C57BL/6小鼠背部,建立不同品系小鼠間皮膚移植免疫排斥模型。將60只模型小鼠隨機(jī)均分為PBS組及低、中、高劑量BMMSC組,分別經(jīng)尾靜脈注射PBS、5×10~4個(gè)BMMSC、10×10~4個(gè)BMMSC、20×10~4個(gè)BMMSC各0.2 m L。記錄各組移植皮膚存活時(shí)間。術(shù)后第7天,HE染色觀察移植皮膚病理組織學(xué)變化;每組隨機(jī)取5只小鼠處死取其脾臟組織及血清,分別用RT-PCR及ELISA法檢測(cè)IL-10、TNF-α、IFN-γ。結(jié)果中、高劑量BMMSC組皮膚存活時(shí)間較PBS組、低劑量BMMSC組長(zhǎng)(P均0.05);中、高劑量BMMSC組皮膚存活時(shí)間比較,P0.05。術(shù)后第7天,病理切片鏡下觀察顯示PBS組炎性細(xì)胞浸潤(rùn)最明顯,高劑量BMMSC組炎性細(xì)胞浸潤(rùn)最少。術(shù)后第7天,與PBS組比較,低、中、高劑量BMMSC組脾臟組織TNF-α、IFN-γmRNA相對(duì)表達(dá)量低(P均0.05)。與PBS組比較,中、高劑量BMMSC組血清IL-10水平高(P均0.05),低、中、高劑量BMMSC組血清TNF-α、IFN-γ水平低(P均0.05);與低劑量BMMSC組比較,中劑量BMMSC組血清TNF-α、IFN-γ水平低(P均0.05),高劑量BMMSC組血清IL-10水平高、TNF-α水平低(P均0.05)。結(jié)論經(jīng)尾靜脈注射適當(dāng)劑量的大鼠BMMSC可延長(zhǎng)不同品系小鼠移植皮膚存活時(shí)間;該作用可能與BMMSC調(diào)控受體小鼠IL-10、TNF-α、IFN-γ的表達(dá)有關(guān)。
[Abstract]:Objective to investigate the effect and mechanism of rat bone marrow mesenchymal stem cell (BMMSC) injection via tail vein on survival time of skin graft in different strains of mice. Methods SD rat BMMSC was isolated and cultured for the third passage. 60 ICR milk samples were obtained by skin sampler. Mouse round skin grafts (12 mm in diameter, transplanted to the back of adult C57BL / 6 mice, An immune rejection model of skin graft was established in different strains of mice. 60 mice were randomly divided into PBS group and low, medium and high dose BMMSC groups. 5 脳 10 ~ 4 BMMSC 10 脳 10 ~ 4 BMMSC were injected into the tail vein respectively. The survival time of the skin grafts in each group was recorded. The histopathological changes of the transplanted skin were observed by HE staining on the 7th day after operation, and the spleen tissues and serum of 5 mice in each group were killed at random. IL-10 TNF- 偽 and IFN- 緯 were detected by RT-PCR and ELISA respectively. The results showed that the skin survival time of high dose BMMSC group was higher than that of PBS group, and that of low dose BMMSC group was 0. 05%, while that of high dose BMMSC group was 0. 05%. The pathological examination showed that the infiltration of inflammatory cells was the most obvious in PBS group and the least in high-dose BMMSC group. On the 7th day after operation, compared with PBS group, the relative expression of TNF- 偽 -IFN- 緯 mRNA in spleen tissue of high dose BMMSC group was lower than that of PBS group (P < 0.05), and compared with PBS group, the relative expression of TNF- 偽 -IFN- 緯 mRNA in spleen tissue of high dose BMMSC group was lower than that of PBS group (P < 0.05). The serum IL-10 levels in the middle and high dose BMMSC groups were 0.05 and 0.05, while those in the middle and high dose BMMSC groups were 0.05 and 0.05, respectively, which were compared with those in the low dose BMMSC group, while in the middle and high dose BMMSC groups, the serum levels of TNF- 偽 and IFN- 緯 were lower than those in the low dose BMMSC group. The serum levels of TNF- 偽 IFN- 緯 in the middle dose BMMSC group were 0.05%, while in the high dose BMMSC group the levels of TNF- 偽 and TNF- 偽 were 0.050.Conclusion injection of appropriate dosage of rat BMMSC via caudal vein can prolong the survival time of skin grafts in different strains of mice. This effect may be related to the regulation of the expression of IL-10, TNF- 偽 and IFN- 緯 by BMMSC.
