電紡絲納米纖維材料負(fù)載凝血酶制備新型止血敷料的可行性研究

【摘要】:任何一種臨床常用的止血材料均存在其獨(dú)特的性能優(yōu)勢(shì),同時(shí)也有各自的局限。探索并研制性能更加優(yōu)良的新型快速止血材料是很有必要的。電紡絲技術(shù)近年來已成為制備藥物遞送系統(tǒng)的一種有效方法。將速效止血藥負(fù)載到電紡絲纖維材料上是一種很有吸引力的制備快速止血材料的途徑。本研究擬探討將速效止血藥凝血酶負(fù)載固定到電紡纖維膜的表面制備高效快速止血材料的可行性及有效性。 目的: 探討以電紡絲納米纖維膜負(fù)載凝血酶制備新型止血敷料的可行性。 方法: 1、篩選合適的高分子材料,經(jīng)電紡絲納米纖維負(fù)載凝血酶制備試驗(yàn)性止血材料; 2、分別測(cè)試試驗(yàn)止血材料在不同溫度、酸堿度及光照條件下的穩(wěn)定性。 3、在不同的動(dòng)物出血模型中與明膠海綿及可吸收止血綾對(duì)比,評(píng)估受試材料的止血性能。 結(jié)果: 以電紡絲膜負(fù)載凝血酶制備止血材料具有可行性。材料在低溫條件下酶活更穩(wěn)定,酸堿環(huán)境及光照條件下易失活。在肝臟滲血模型中試驗(yàn)材料的止血效果優(yōu)于明膠海綿。在動(dòng)脈搏動(dòng)性出血模型中其止血可靠性優(yōu)于可吸收止血綾。在大動(dòng)脈縫合修補(bǔ)后滲血模型中試驗(yàn)材料的止血效果不及可吸收止血綾。 結(jié)論: 電紡絲膜負(fù)載凝血酶制備的新型止血材料有著良好的臨床應(yīng)用前景。
[Abstract]:Any kind of hemostatic material commonly used in clinic has its unique performance advantages, but also has its own limitations. It is necessary to explore and develop a new type of rapid hemostatic material with better properties. Electrospinning technology has become an effective method to prepare drug delivery system in recent years. Loading quick hemostatic drugs on electrospun fiber materials is an attractive way to prepare rapid hemostatic materials. The purpose of this study was to investigate the feasibility and effectiveness of immobilization of available hemostatic drug thrombin on the surface of electrospun fiber membrane to prepare efficient and rapid hemostatic materials. Objective: to investigate the feasibility of preparing a new hemostatic dressing with thrombin loaded with electrospun nanofiber membrane. Methods: 1. The suitable polymer materials were selected and the experimental hemostatic materials were prepared by electrospun nanofiber loaded with thrombin. 2. The stability of the experimental hemostatic materials under different temperature, pH and light conditions was tested respectively. 3. The hemostatic properties of the tested materials were evaluated by comparing with gelatin sponge and absorbable hemostatic silk in different animal bleeding models. Results: it is feasible to prepare hemostatic material by electrospinning membrane loaded thrombin. The enzyme activity of the material was more stable at low temperature, and it was easy to be inactivated in acid-base environment and light. The hemostatic effect of the experimental material is better than that of gelatin sponge in the model of liver bleeding. The hemostatic reliability of arterial pulsatile hemorrhage model is better than that of absorbable hemostatic model. The hemostatic effect of the experimental material in the bleeding model after artery suture repair was not as good as that of absorbable hemostasis. Conclusion: the new hemostatic material prepared by electrospinning membrane loaded with thrombin has a good clinical application prospect.
