Lys調節(jié)PLGA納米纖維的體外降解產物酸度的研究

【摘要】:采用向PLGA納米纖維中添加堿性氨基酸——賴氨酸(Lys)的方法來降低降解產物的集酸程度.用電紡絲技術制備載Lys的PLGA/Lys復合納米纖維,并用SEM、DSC、及力學測試等表征手段對該復合納米纖維的形貌、熱學性質以及力學性質等方面進行表征;研究PLGA/Lys復合納米纖維中Lys的釋放特性;對PLGA與PLGA/Lys納米纖維進行體外降解實驗,評價纖維降解過程中的形貌和降解液pH值;用已降解8周的納米纖維降解液對血管平滑肌細胞進行培養(yǎng),MTT檢測細胞毒性.結果表明,在電壓為12 kV,注射速率1.0mL/h,溫度23℃,濕度20%,接收距離14 cm時可以紡出形貌較好的PLGA/Lys復合納米纖維,Lys含量越多,復合納米纖維的直徑越細.納米纖維中Lys的含量影響Lys的釋放特性.Lys促進PLGA的降解并對改善其降解產物的酸度有一定的緩解作用,含2%Lys的納米纖維降解8周后可以使降解液pH維持在7.4~6.7之間.降解的酸性產物不利于平滑肌細胞生長,但含Lys降解液的培養(yǎng)基中細胞生長狀態(tài)良好,這可能是由于Lys能減輕酸性降解產物對細胞功能的影響,同時也能為細胞增殖提供營養(yǎng)物質.
[Abstract]:The alkaline amino acid-lysine (Lys) was added to the PLGA nanofibers to reduce the acid concentration of the degradation products. PLGA/Lys composite nanofibers loaded with Lys were prepared by electrospinning technique. The morphology, thermal properties and mechanical properties of the composite nanofibers were characterized by SEM,DSC, and mechanical tests. The release characteristics of Lys from PLGA/Lys composite nanofibers were studied, and the degradation experiments of PLGA and PLGA/Lys nanofibers in vitro were carried out to evaluate the morphology and pH value of the degradation solution. Vascular smooth muscle cells (VSMCs) were cultured with biodegradable nanofiber solution for 8 weeks. The cytotoxicity of vascular smooth muscle cells (VSMCs) was detected by MTT.
