機(jī)械力及力學(xué)信號(hào)轉(zhuǎn)導(dǎo)影響干細(xì)胞命運(yùn)的研究進(jìn)展

【摘要】:干細(xì)胞作為一種未分化的祖細(xì)胞,目前已被廣泛應(yīng)用于開展組織損傷修復(fù)、再生以及干細(xì)胞特異譜系分化的研究.大量研究表明,干細(xì)胞所處的微環(huán)境對(duì)調(diào)控干細(xì)胞的生長(zhǎng)和分化具有重要作用,多種溶液介質(zhì)、細(xì)胞外基質(zhì)和信號(hào)通路等參與了干細(xì)胞命運(yùn)的調(diào)控.盡管已有大量研究證明,溶液介質(zhì)(如激素和生長(zhǎng)因子)在干細(xì)胞的生長(zhǎng)和分化中發(fā)揮重要作用,但近年來越來越多的研究表明,機(jī)械力及力學(xué)信號(hào)轉(zhuǎn)導(dǎo)同樣在干細(xì)胞自我更新、分化、衰老和凋亡等細(xì)胞生理過程中起到重要的作用.本文將對(duì)機(jī)械應(yīng)力響應(yīng)的細(xì)胞基礎(chǔ)、生物力學(xué)及力學(xué)信號(hào)調(diào)控干細(xì)胞自我更新和分化,以及生物力學(xué)調(diào)控干細(xì)胞命運(yùn)可能的作用機(jī)制幾個(gè)方面加以綜述.
[Abstract]:As a kind of undifferentiated progenitor cells, stem cells have been widely used in tissue damage repair, regeneration and stem cell specific lineage differentiation. A large number of studies have shown that the microenvironment in which stem cells are located plays an important role in regulating the growth and differentiation of stem cells, and many kinds of solution media, extracellular matrix and signal pathway are involved in the regulation of stem cell fate. Although a large number of studies have shown that solution media (such as hormones and growth factors) play an important role in the growth and differentiation of stem cells, the more recent studies have shown that Mechanical and mechanical signal transduction also play an important role in the physiological processes of stem cells, such as self-renewal, differentiation, senescence and apoptosis. This article reviews the cellular basis of mechanical stress response, the self-renewal and differentiation of stem cells regulated by biomechanical and mechanical signals, and the possible mechanism of biomechanical regulation of stem cell fate.
