脂肪干細胞與黃芪多糖膠原的生物相容性及其復(fù)合材料促進血管新生效果的實驗研究

【摘要】:研究目的: 1研究脂肪干細胞和黃芪多糖膠原復(fù)合材料的可行性和生物相容性。 2研究脂肪干細胞聯(lián)合黃芪多糖膠原對血管新生的影響。 3探討黃芪多糖膠原與脂肪干細胞之間的作用影響及可能存在的機制,評價該復(fù)合物的優(yōu)劣、目前存在的問題以及在后續(xù)實驗及臨床研究中的意義 研究方法: 1體外培養(yǎng)脂肪干細胞,觀察其生物學(xué)特性。 2制備黃芪多糖膠原,檢測膠原相關(guān)特性。 3將脂肪干細胞種植入黃芪多糖膠原,通過光鏡觀察、黏附率檢測、細胞增殖等手段研究脂肪干細胞與黃芪多糖膠原的生物相容性。 4ELISA法檢測共培養(yǎng)后,脂肪干細胞與黃芪多糖膠原復(fù)合物所分泌的VEGF含量。 5雞胚尿囊膜實驗檢測脂肪干細胞與黃芪多糖膠原復(fù)合物在促進血管新生方面的效果。 研究結(jié)果: 1脂肪干細胞的生物學(xué)研究:培養(yǎng)的脂肪干細胞形態(tài)良好,生長良好。 2脂肪干細胞與黃芪多糖膠原的生物相容性:脂肪干細胞可以在黃芪多糖膠原支架上黏附、生長和增殖,其黏附率為:%;脂肪干細胞在黃芪多糖膠原支架上能夠正常生長和增殖,并且黃芪多糖膠原在一定的階段內(nèi)對脂肪干細胞的增殖有促進作用。 3黃芪多糖膠原和脂肪干細胞共培養(yǎng)時,對脂肪干細胞所分泌的細胞因子VEGF有促進作用。 4脂肪干細胞與黃芪多糖膠原復(fù)合物對雞胚尿囊膜血管新生具有促進作用,并且明顯高于對照組。 研究結(jié)論: 脂肪干細胞與黃芪多糖膠原具有良好的生物相容性,黃芪多糖膠原可以成為脂肪干細胞一個理想的支架載體。該復(fù)合材料具有促進血管新生的作用,其機制可能與脂肪干細胞分泌的細胞因子以及脂肪干細胞的橫向分化相關(guān)。對于這方面可以有進一步的后續(xù)研究。
[Abstract]:Objectives of the study: 1 to study the feasibility and biocompatibility of the composite of Astragalus polysaccharide collagen and adipose stem cells. 2 to study the effect of adipose stem cells combined with Astragalus polysaccharide collagen on angiogenesis. 3 to explore the effect of Astragalus polysaccharides collagen on adipose stem cells and its possible mechanism, to evaluate the advantages and disadvantages of the complex, the existing problems and its significance in subsequent experiments and clinical studies Research methods: 1 Adipose stem cells were cultured in vitro and their biological characteristics were observed. 2 Astragalus polysaccharides collagen was prepared and the characteristics of collagen were detected. (3) Astragalus polysaccharide collagen was implanted into adipose stem cells. The biocompatibility of adipose stem cells with Astragalus polysaccharide collagen was studied by means of light microscope, adhesion test and cell proliferation. The content of VEGF secreted by Astragalus polysaccharides collagen complex was measured by 4ELISA method. 5 chicken embryo allantoic membrane test was used to detect the effect of adipose stem cells and Astragalus polysaccharides collagen complex on promoting angiogenesis. Results of the study: 1 Biological study of adipose stem cells: the cultured adipose stem cells have good morphology and good growth. 2 biocompatibility of Astragalus polysaccharide collagen with adipose stem cells: adipose stem cells can adhere, grow and proliferate on the scaffold of Astragalus polysaccharides collagen, and the adhesion rate of adipose stem cells is 10%, and adipose stem cells can grow and proliferate normally on the scaffold of Astragalus polysaccharide collagen. And Astragalus polysaccharides collagen can promote the proliferation of adipose stem cells at a certain stage. 3 Astragalus polysaccharides collagen and adipose stem cells co-cultured can promote the cytokine VEGF secreted by adipose stem cells. 4Adipose stem cells and Astragalus polysaccharides collagen complex could promote angiogenesis of chicken embryo allantoic membrane, and it was significantly higher than that of control group. The study concluded that: Astragalus polysaccharide collagen has good biocompatibility with adipose stem cells, and it can be an ideal scaffold carrier for adipose stem cells. The composite has the effect of promoting angiogenesis, and its mechanism may be related to cytokines secreted by adipose stem cells and lateral differentiation of adipose stem cells. There can be further research in this area.
