醫(yī)用多孔鉭材料復合大鼠軟骨細胞的生長特性及功能變化

【摘要】:目的評價多孔鉭材料復合大鼠軟骨細胞的體外生長特性及功能變化。方法選取3周齡SD大鼠,分離關節(jié)軟骨細胞并進行體外擴增培養(yǎng),取生長狀態(tài)良好的第2代細胞,鑒定后以1×106/ml密度接種于多孔鉭上進行復合培養(yǎng)。采用倒置相差顯微鏡、掃描電鏡、透射電鏡觀察軟骨細胞在多孔鉭上的形態(tài)特點,比色法測定復合培養(yǎng)的軟骨細胞中糖胺多糖(GAG)的含量。結果經(jīng)Ⅱ型膠原免疫細胞化學染色、甲苯胺藍染色及番紅O染色鑒定所獲細胞為軟骨細胞。倒置相差顯微鏡觀察可見多孔鉭邊緣及培養(yǎng)板底部黏附大量軟骨細胞;掃描電鏡觀察可見軟骨細胞在多孔鉭表面及孔隙內黏附、伸展、增殖,并分泌細胞外基質;透射電鏡觀察顯示軟骨細胞內超微結構大小及形態(tài)無明顯改變;比色法檢測顯示軟骨細胞在多孔鉭支架上可持續(xù)分泌GAG。結論多孔鉭材料與軟骨細胞體外共培養(yǎng)顯示出良好的生物相容性,適合作為軟骨組織工程的支架材料。
[Abstract]:Objective to evaluate the growth characteristics and functional changes of porous tantalum composite chondrocytes in vitro.Methods the articular chondrocytes were isolated from 3-week-old SD rats and cultured in vitro. The second generation cells in good growth state were identified and cultured on porous tantalum with a density of 1 脳 106/ml.The morphologic characteristics of chondrocytes on porous tantalum were observed by inverted phase contrast microscope, scanning electron microscope and transmission electron microscope.
